In the invention, penicillium purpurogenum separated and sieved from coast intertidal zone sludge is adopted to prepare the seawater microbial flocculants; potato juice is taken as raw materials to prepare fermentation medium; penicillium purpurogenum EL-O2 strains are inoculated in the fermentation medium, and are cultured at 25-35DEG C at shaker rotating speed of 160-200rpm to obtain ferment culture solution; the ferment culture solution is centrifugalized at a high speed under the conditions that the temperature is 4-8DGE C and the rotating speed is 8000-12000rpm to obtain ferment liquid supernatant; the ferment liquid supernatant is precipitated with ethanol and is centrifugalized at a high speed at 4-8DEG C and at the rotating speed of 10000-12000rpm to obtain precipitate; and the precipitate is subject to vacuum freeze-drying under the conditions that the temperature is 45DEG C below zero to 20DEG C below zero and the vacuum degree is lower than 15pa to obtain the finished seawater microbial flocculants.
