Embodiment Construction

[0026] 1: Processing milk samples (milk sample I, milk sample II, milk sample III), 2 mg milk powder or 2 mL liquid milk is first dissolved with secondary water to the total volume of 50 mL, and then diluted 10 times and then used 10 KD ultra Filter membrane ultrafiltration, after centrifugation at 20000 rpm for 30 min, take the supernatant and dilute 10000 times.

[0027] 2: Take 30 μL 1.0 × 10 -5 The M melamine solution (37.8 ng) was added to 1.0 mL of the above-mentioned ultrafiltration diluted milk sample.

[0028] Add AuNPs and DNA polyT with a molar ratio of 1:23 to a 1.5 mL centrifuge tube 55 (The final concentration of AuNPs of gold nanoparticles is 2nM, DNA polyT 55 The final concentration is 0.046uM), vortex and mix well, and react at room temperature for 5-10 min. Then add it to the mixed solution of milk and melamine, and detect by PRLS method. When measuring the scattering spectrum, the excitation wavelength and emission wavelength are equal, that is em = ex .

Abstract

The invention relates to a quick detection method for melamine in dairy products. Based on the principle of three intermolecular hydrogen bonds between melamine and thymine, the method is set up for quickly and flexibly detecting melamine in dairy products under an aqueous solution buffer system by the aid of gold nano particles wound by polyT55. The specific hydrogen bonds between the melamine and the polyT55 enable the gold nano particles wound by the polyT55 to be aggregated so as to cause variation of PRLS (prolactin signal) of gold size, and detection of melamine in dairy products is realized. The method is ingenious in design, high in selectivity and flexibility, free of modification, simple in operation and free of large instruments.

Description

Technical field [0001] The invention relates to a detection method of melamine. Background technique [0002] Melamine, due to its nitrogen content as high as 66%, is often used by illegal traders to mix it in milk or animal feed to increase protein content. This illegally added melamine in milk or animal feed has caused kidney stones and even deaths in infants. Therefore, people are paying close attention to melamine testing recently. [0003] The current methods for detecting melamine mainly include high performance liquid method (HPLC), high performance liquid phase coupled with mass spectrometry (HPLC-MS) and other methods. However, these methods involve large instruments and are not conducive to rapid real-time analysis and detection. In view of the current status of existing detection methods, it is of great significance to develop a new method for simple, sensitive and rapid detection of melamine. Summary of the invention [0004] The purpose of the present invention is to ...
