Example Embodiment

[0018] The present invention will be described in detail through the following examples:

[0019] The industrial production method of the lipopeptide biosurfactant includes the following steps:

[0020] (1) Bacterial breeding: The starting bacterium was selected from the Bacillus subtilis No. 10366 of China Industrial Microorganism Preservation and Management Center. After enrichment culture, blood plate separation and screening, a high-yield production strain was obtained, No. VTS-1106 ;

[0021] (2) Inclined seed cultivation: the ratio of the medium is 20g / L glucose, 5g / L ammonium nitrate, 1.0g / L potassium dihydrogen phosphate, 2g / L disodium hydrogen phosphate, 0.5g / L magnesium sulfate, yeast powder 0.8g / L, L-glutamic acid 0.8g / L, niacin 0.0002g / L, the balance is water; pH value 7.2-7.3, 0.1Mpa sterilization for 20min, slant culture at 37℃ constant temperature for 24h, check if there is no Infect the bacteria for later use. The total amount of slant culture is calculated as ≥ 6%...

Abstract

The invention relates to an industrial production method of a dry rat glycolipid bio-surfactant powder. The method comprises the following steps of: (1) breeding a strain; (2) slantly culturing the strain; (3) carrying out enlarged culture on the strain; (4) industrially fermenting and culturing the strain; and (5) centrifuging a produced lipopeptide bio-surfactant fermentation liquor by a high speed centrifugal machine at a rotation speed of 8000 rpm/min, so as to respectively obtain a lipopeptide separating liquor and bacillus subtilis. The surface tension of the stoste of the lipopeptide bio-surfactant fermentation liquor produced with the method provided by the invention is less than 26 mN/m; and after being centrifuged to remove thalli and then diluted by 500 times, the stoste of the lipopeptide bio-surfactant fermentation liquor obtains the surface tension of less than 34 mN/m. The separated lipopeptide separating liquor can be used for ternary complex flooding oil-extraction; and the flooding effect of the lipopeptide separating liquor can be improved by 8-16% on the basis of water flooding. It is detected that the bacterial count of the separated bacillus subtilis can reach more than 10 billions in every 6 ml of the fermentation liquor; and the bacillus subtilis can be used for producing and processing EM (E Mycin) bacterial liquids and is applied to the fields, such as agriculture, livestock breeding, and sewage treatment (industrial sewage and breeding sewage).

Description

Technical field: [0001] The invention relates to an industrial production method of a lipopeptide biosurfactant. Background technique: [0002] Surfactant refers to a substance with a certain structure and hydrophilic and lipophilic properties that can significantly reduce surface tension and interfacial tension. It is mainly divided into chemical surfactants and biological surfactants. The most commonly used ones are chemical surfactants, but they are generally toxic, harmful and pollute the environment during their production and use. In contrast, biological surfactants are mainly produced by the physiological metabolism of microorganisms. In addition to good surface activity, they also have structural diversity, low toxicity or non-toxicity, biodegradability, and adapt to extreme temperatures, pH and salt characteristics such as degree. Its raw materials are generally derived from plants instead of petroleum, and it has the characteristics of low cost, harmless producti...
