According to the lignin degradation solution and the methods disclosed by the invention, the synergistic oxidative degradation of macromolecular lignin with rich structural diversity is realized, and compared with a degradation reaction system with single ligninase, the degradation rate of the macromolecules of the lignin can each 30-50%, the degradation conversion efficiency is significantly improved, and the degradation solution and the methods can be applied to the fields of bio-refinery of lignocelluloses, biological pulping or environmental treatment and the like.

The invention relates to a method for studying the structural diversity of a daqu bacterial community, i.e. the denaturing gradient gel electrophoresis (DGGE) technology, which belongs to the technical field of microbial ecology and mainly comprises the following steps that: 1), daqu genomic deoxyribonucleic acid (DNA) is directly extracted; 2), bacterial universal primers are selected, and specific DNA fragments in the bacterial ribosome DNA are amplified; 3), polymerase chain reaction (PCR) products are separated through DGGE; 4), corresponding strips of microbes in the DGGE fingerprint map are recovered in a gel cutting way; 5), PCR is carried out again, products are connected to a T carrier, the blue and white spot screening is carried out, and positive clone verification is carried out; and 6), the species information of the corresponding microbes of the DGGE strips is obtained through the sequence test. The method does not rely on the molecular biology technology of the traditional culture method, has the characteristics of sensitivity, convenience and accuracy and solves the difficult problems to study some microbes incapable of being cultured or difficult to culture, and the method provides the technical basis for the verification of the daqu bacterial community structure and the discovery of new wine brewing microbes or functional microbes.

The invention discloses a method for extracting total DNAs of soil microorganisms at high purity. CTAB (Cetyltrimethyl Ammonium Bromide), lysozyme, protease and SDS (Sodium Dodecyl Sulfonate) act together to perform lysis of cells and simultaneously freeze and thaw the cells repeatedly; therefore, an excellent cell lysis effect is achieved; the humic acid is removed by utilizing PVP (Polyvinyl Pyrrolidone), CaC12 and BSA (Bull Serum Albumin); and the DNAs are precipitated by utilizing isopropanol so that the purity of the obtained DNAs is high. The method is simple and convenient; and without being purified, the DNAs obtained by the method can be used for subsequent PCR (Polymerase Chain Reaction) analysis and DGGE (Denaturing Gradient Gel Electrophoresis) analysis. The purity of the extracted DNAs meets the requirement for directly performing subsequent molecular biological research; the extracted DNA segments can be subjected to 16SrDNA amplification and DGGE atlas analysis; glue cutting recovery and basic group sequencing can be performed on specific stripes of the DGGE band; unknown bacteria can be identified through comparison with the conventional sequences in an RDP (Ribosomal Database Project) database or by establishing a new sequence probe; and therefore, the structural composition of the microbial community can be determined, and a foundation can be laid for researching the structural diversity and ecological functions of the soil microorganisms in the rhizosphere of the mulberry in future.

The invention discloses a method for extracting total DNA (deoxyribonucleic acid) of mulberry rhizosphere soil microorganisms by adopting a plurality of measures, which can achieve a good cell lysis effect by utilizing PVP (polyvinyl pyrrolidone) prewashing to remove humic acid, the synergy of CTAB (cetyltrimethyl ammonium bromide), lysozyme, protease and SDS (sodium dodecyl sulfate) to lyse cells and repetitively freezing and thawing the cells at the same time and utilizes PEG8000 to precipitate DNA, so that the purity of the obtained DNA is high. The adopted method is simple and convenient, and the DNA can be used in subsequent PCR (Polymerase Chain Reaction) analysis and DGGE (denaturing gradient gel electrophoresis) analysis without needing to be purified. The extracted DNA is so pure as to meet the requirement of direct subsequent molecular biology research, an extracted DNA fragment can be used in the amplification and DGGE map analysis of 16SrDNA, moreover, the specific band gel extraction and base sequencing of a DGGE band can be carried out, comparison with the sequence existing in an RDP (ribosomal database project) database can be carried out, or a new sequence probe can be established to identify unknown bacteria, so that the composition of a microbial community structure can be determined, and thereby laying a foundation for the future research of the community structure diversity and the ecological functions of the mulberry rhizosphere soil microorganisms.

Bacillus cereus and application thereof to improve plant drought-resistant capability

The invention discloses a strain of bacillus cereus which is preserved in China General Microbiology Center of Committee for Culture Collection of Microorganisms since 31st December, 2014, and the preservation number is CGMCC No. 7069. The bacillus cereus L90 provided by the invention is capable of improving the drought resistant capability of juglans regia. Experiment research shows that after the bacillus cereus provided by the invention is inoculated to the surround of juglans regia roots and juglans regia is stressed by drought for a certain time, juglans regia is substantially improved in bacterial community structure diversity in rhizosphere soil compared with juglans regia inoculated with common growth-promoting bacteria, the leaf relative water content and the water utilization efficiency are improved, and the drought resistance of the plant is enhanced. Therefore, the bacillus cereus provided by the invention or a bacterium preparation by taking bacillus cereus as a raw material can be used as an inoculation agent and applied to drought environment, and the bacillus cereus provided by the invention helps to provides an excellent bacterial strain resource for improving the drought resistance of juglans regia.
