The preparation method comprises the following steps: degumming cocoon layers in a Na2CO3 boiling water solution for twice, then flushing by using deionized water, wringing, and drying the cocoon layers in a drying oven; putting silk fibroin fibers in an LiBr solution, performing vibration treatment, filtering, then feeding into a dialysis bag, and then concentrating a solution; adding EDC powder into a silk fibroin solution, stirring, completely dissolving, and uniformly mixing; and spreading a mixed solution on a polyethylene plate, naturally drying at normal temperature, or drying the mixed solution in the drying oven to prepare a silk fibroin membrane. The preparation method disclosed by the invention is simple to operate and mild in reaction condition; and meanwhile, other high-molecular polymers are not introduced, the silk fibroin membrane which is water-insoluble and is flexible in a dry state can be prepared, the silk fibroin membrane is expected to be applied to the fields of medicines and health, tissue engineering and the like, application routes of silk fibroin can be expanded, and a novel method for preparing high-molecular protein membranes with excellent performance can also be provided.
