Problems solved by technology

[0003] Existing methods for the separation and purification of monascus red pigment include column chromatography, thin plate chromatography, liquid chromatography, etc. Different substances, but homologous pigments, orange pigments, yellow pigments, amino acids and protein substances that have little difference in hydrophilicity with red pigments are difficult to separate
Moreover, purification by chromatography requires a large amount of organic solvents, chromatography materials and expensive instruments and equipment, which limits its application in industrialization due to high cost

Abstract

The invention discloses a purifying method of hydroxy-containing pigment; the technology thereof is simple, the cost is low, and suitable for industrial production. The method comprises the following steps: pasting and high-pressure processing a starch emulsion to obtain a starch paste, uniformly mixing the obtained starch paste with a hydroxy-containing pigment water solution, aging to obtain a pigment-containing colloidal retrogradation starch; adding a high-temperature amylase into the pigment-containing colloidal retrogradation starch to enzymatically hydrolyzing the starch which is not participated in the retrogradation, separating and discarding the supernatant, collecting the sediment, washing to obtain a purified hydroxy pigment contained retrogradation starch; dissolving the purified hydroxy pigment contained retrogradation starch into a dense alkali liquor to open the hydrogen bond of the starch and make the starch into a water-soluble starch; releasing the pigment containing hydroxy, regulating the pH value thereof to 7.0 in order to neutralize the alkali liquor into a salt; using ethyl alcohol to subside the starch therein, collecting the solution containing the hydroxy pigment; concentrating and drying the obtained solution into a solid; processing the solid by vibration, sieving the separate the block crystal therein, thus the lower layer powder is the purified pigment powder. The method removes the impurities by containing hydroxy; and the method is suitable for industrial production.

Description

technical field [0001] The invention relates to the field of food biotechnology, in particular to a method for purifying hydroxyl-containing pigments. Background technique [0002] Monascus red pigment is a pigment produced during the fermentation process of Monascus sp. At present, the company mainly adopts the method of liquid fermentation of rice flour and spray drying of soybean protein in the later stage. Such products contain a large amount of water-soluble carbohydrates, polypeptides, proteins and other substances that are not utilized by Monascus, which reduces the purity of the pigment, increases the amount of Monascus red pigment added in food, and reduces the stability of the product. [0003] Existing methods for the separation and purification of monascus red pigment include column chromatography, thin plate chromatography, liquid chromatography, etc. It is difficult to separate homologous pigments, orange pigments, yellow pigments, amino acids and protein sub...
