Abstract

The invention provides a gene engineering method for improving age-related senescence of mesenchymal stem cells of aged rats. The method comprises the following steps: putting passage-stable mesenchymal stem cells obtained from marrow of the aged rats in a conventional culture medium provided with TPP1 overexpression slow virus particles (MOI is 100), infecting for 24 hours, and then replacing the culture medium into the conventional culture medium, adding puromycin in concentration of 5 microgrammes per microliter after 3 days, screening for 7 days, and applying the screened mesenchymal stem cells to cell transplantation therapy. The method has the beneficial effects that through overexpression of TPP1 in the mesenchymal stem cells of the aged rats, the age-related senescence of the mesenchymal stem cells of the aged rats can be obviously improved, so that the purpose of improving the effect of treating ischemic myocardial diseases can be achieved.
