Abstract

The invention discloses a method for preparing major yolk protein from a coelomic fluid of sea cucumber. The specific processes comprise the steps of pretreatment of the coelomic fluid of the sea cucumber, low-temperature desalting by a dialysis method, extraction of the major yolk protein, redissolving, dialysis and drying. The by-product, namely the coelomic fluid of the sea cucumber, generatedduring processing of the sea cucumber is used as a raw material, so that the utilization rate of the by-product generated during processing of the sea cucumber is raised, the protein resource of the coelomic fluid of the sea cucumber is fully developed and utilized, meanwhile, the pollution of the by-product of the sea cucumber to the environment is reduced, and the method has great significance to development and utilization of the major yolk protein in the coelomic fluid of the sea cucumber. Compared with traditional protein precipitation methods, which are hard to achieve scale production,such as ammonium sulfate salting-out, ethanol precipitation and isoelectric precipitation, the method provided by the invention is mild in operation condition, and therefore denaturation of biomacromolecules is not liable to occur; the extraction rate is up to 70%; and the method provided by the invention is simple, meanwhile, a large number of samples can be treated at a time, assistance of complex instruments is not needed, and the method is suitable for large-scale production.
