[0020] 1. Cloning, sequence analysis and evolutionary tree analysis of Pm21 candidate gene DvRGA-1

[0021] The present invention uses long-distance PCR technology, using P1 (5'-GAAATAATGGGCACCCCTTCTTA CTA-3') and P2 (5'-ATGCACTTACATCGTTAGTTACAACATC-3') as primers, and disease-resistant tufted wheat from the Cambridge Botanical Garden in England (known and public, Chen et al. Development and molecular cytogenetic analysis of wheat-Haynaldia villosa 6VS / 6AL translocation lines specifying resistance topowdery mildew. Theor Appl Genet. 1995,91:1125-1128) cloned a gene in the genome, because the gene is derived from the ( D asyprum v illosum), and is an analogue of disease resistance genes ( R esistance G ene A nalog), so it was named DvRGA-1. DNA sequencing found that the full length of the genomic DNA of the DvRGA-1 gene is 3699bp, and the specific sequence is SEQ ID NO. 1. The DvRGA-1 gene has three exons and two introns (see figure 1 ), RT-PCR analysis revealed that the cDNA sequ...

Abstract

The invention discloses a haynaldia villosa powdery mildew resistant gene DvRGA-1, DvRGA-2 and the application thereof, and belongs to the field of gene engineering. The genomic DNA sequence of haynaldia villosa DvRGA-1 is SEQ ID NO. 1, the cDNA sequence is SEQ ID NO. 2, and the amino acid sequence is SEQ ID NO. 3. The genomic DNA sequence of DvRGA-2 is SEQ ID No. 4, the cDNA sequence is SEQ ID NO. 5, and the amino acid sequence is SEQ ID NO. 6. The DvRGA-1 is located at the Pm21 locus of haynaldia villosa powdery mildew resistant gene. Gene silencing analysis shows that the silencing of DvRGA-1 and DvRGA-2 can lead to complete powdery mildew of disease-resistant wheat; the analysis of transient expression shows that DvRGA-1 and DvRGA-2 can significantly reduce the haustorium index of powdery mildew of susceptible wheat. Therefore, the haynaldia villosa DvRGA-1 and DvRGA-2 genes and the coded proteins have important application value in wheat powdery mildew resistant breeding.

Description

Technical field [0001] The present invention discloses two powdery mildew resistance genes DvRGA-1 and DvRGA-2 of Dasyprum villosum, both of which are disease resistance gene analogs (RGA), which encode typical spiral coil-nucleotide binding sites- Disease-resistant proteins rich in leucine repeats (CC-NBS-LRR) can improve the resistance of wheat to powdery mildew and belong to the field of genetic engineering. Background technique [0002] Wheat is one of the main food crops in the world, and its production is threatened by various diseases. Wheat powdery mildew is a fungal disease caused by wheat powdery mildew (Blumeria graminis f.sp.tritici), which is an important factor leading to reduced wheat production. An economical and effective way to control the harm of wheat powdery mildew is to explore disease-resistant resources and cultivate disease-resistant varieties. More than 100 wheat powdery mildew resistance genes have been reported (McIntosh et al. Catalogue of gene symb...
