Problems solved by technology

However, currently known biocontrol agents for inhibiting bacterial diseases still have short shelf life, are not resistant to salt, are not resistant to osmotic pressure, chemical bacteriostatic agents have inactivation effects on it, cannot be shared with chemical bacteriostatic agents, have poor stress resistance and Disadvantages such as poor control effect and slow effect during use
[0004] At present, the country vigorously advocates ecological planting and the development of sustainable green agriculture. Therefore, some microbial biocontrol agents have emerged as the times require in the prevention and treatment of citrus diseases. Poor resistance, weak ability to determine the value of plants, unable to form the proper amount of antagonistic bacteria, thus weak ability to inhibit pathogenic bacteria, and unstable control effect
Most bacillus are sensitive to chemical sterilization and antibacterial agents, and cannot be used at the same time. Some chemical agents used to prevent and control diseases have a greater impact on the growth and reproduction of biocontrol bacteria
Therefore, microbial biocontrol agents are subject to various restrictions in the process of disease control.

Embodiment 1

[0034] Step 1: Bacillus (named Bacillus DY-3) is subjected to strain rejuvenation and activation treatment in LB medium:

[0035] Step S1: taking the Bacillus strains in a preserved state for use;

[0036] Step S2: After the LB medium is sterilized by a conventional high-temperature sterilizer, pour the LB medium into a 9 cm sterile petri dish on an ultra-clean workbench, about 15 mL per petri dish;

[0037] Step S3: After the LB medium is solidified, use an inoculation loop to pick up the preserved Bacillus species lawn and scratch the board;

[0038] Step S4: place it horizontally for more than half an hour, and wait for the bacterial lawn to be adsorbed on the LB medium;

[0039] Step S5: place the plate upside down in a 37°C incubator and incubate for 30 hours;

[0040] Step S6: Observe the cultured Bacillus strains, select strong colonies, select some of the colonies and inoculate them into new LB medium for culture, repeat 2-3 times, so as to obtain well-growing strain...

Abstract

The invention provides a microbial biocontrol agent for inhibiting bacterial diseases and a preparation method thereof. The microbial biocontrol agent is prepared from the following components in parts by mass: 2-6 parts of bacillus preparation and 1 part of ethylicin. During preparation, a bacillus is subjected to strain rejuvenation and activating treatment in an LB culture medium; a triangularflask culture medium, a seeding tank culture medium and a fermenter culture medium are separately disinfected; a strain is inoculated into the triangular flask culture medium for culture for 20h; an obtained strain is inoculated into the seeding tank culture medium for culture for 16h; then an obtained strain is inoculated into the fermenter culture medium for culture for 28h to obtain the bacillus preparation; and the bacillus preparation is mixed with ethylicin in proportion to obtain the microbial biocontrol agent for inhibiting the bacterial diseases. The microbial biocontrol agent has theadvantages of convenient use, time and labor saving, use by being mixed with a liquid fertilizer and antibiotics, no influence on the activity of a microbial agent and good comprehensive benefits.

Description

technical field [0001] The invention belongs to the technical field of prevention and treatment of citrus diseases, and in particular relates to a microbial biocontrol agent used in combination with bacillus and a chemical antibacterial agent for inhibiting bacterial diseases and a preparation method thereof. Background technique [0002] The antibacterial spectrum, antibacterial effect and mechanism of different biocontrol strains are quite different. Biocontrol Bacillus has been found in many tests to have the ability to inhibit a variety of pathogenic bacteria that exist widely in nature, and is more friendly to the environment and humans. Therefore, it is favored by researchers in plant protection and environmental protection at home and abroad. [0003] At present, a lot of research has been done on Bacillus with biocontrol function in our country. And the research on the application of Bacillus subtilis to control plant diseases has reached the advanced level in the w...
