Problems solved by technology

The main disadvantages of this method are: 1. It is difficult to distinguish protein subunits with similar molecular weights, and there is such a problem in the identification of lipoxygenase deletion. Since lipoxygenase is composed of three isoenzymes, the molecular weight is between 94-97Kd It is relatively easy to identify the simultaneous deletion of three isoenzymes, but it is difficult to identify the absence of one of the enzymes, which has the disadvantage of high misjudgment rate; 2. The detection cycle is long, and it takes at least 10 more than an hour
3. The biggest disadvantage: pre-emergence sampling inspection, damage to seeds, unable to ensure safe emergence of seeds containing target genes, and reproduction of offspring
The main disadvantages of this method are as follows: 1. The operating procedures are complicated and difficult to grasp
2. There are also the problems of long detection period and low work efficiency. 3. Like the SDS-PAGE identification method, there are also problems of pre-emergence sampling detection, damage to seeds, and failure to ensure safe emergence of seeds containing target genes and reproduction of offspring.

Abstract

The invention discloses a method for identifying lipoxygenase deficiency of a soybean variety. The method comprises the steps of (1) sowing a soybean seed to be identified and taking cotyledons growing after seedling emergence as samples to extract proteins, (2) carrying out electrophoresis on the extracted proteins by adopting an ISDS-PAGE method and analyzing and identifying whether three targetbands Lox-1, 2 and 3 are missing or not. According to the invention, seed sampling before sowing is changed into sampling after sowing and seedling emergence, therefore, the seed is not damaged, theprobability that the seed is infected with plant diseases and insect pests is reduced, the safe seedling emergence of the soybean seed containing target characters can be ensured, the death rate of individuals containing target gene characters is greatly reduced, and the survival rate of the individuals containing the target characters is increased to 99% or above from about 50% of that of a traditional sampling method. By means of the identification method, the survival rate of the shape containing the target gene is greatly increased, and time and labor cost is effectively saved.

Description

technical field [0001] The invention relates to a method for identifying lipoxygenase deficiency in soybean varieties, in particular to a method for identifying lipoxygenase deficiency in soybean varieties by using improved SDS-PAGE, and belongs to the field of identification of lipoxygenase-deficient soybean varieties. Background technique [0002] Lipoxygenase (lipoxygenase, abbreviated: Lox) referred to as lipoxygenase, also known as lipoxygenase, includes three isozymes, namely: Lox-1, Lox-2 and Lox-3. When soybean seeds are broken and exposed to oxygen, the unsaturated fatty acids such as linoleic acid and linolenic acid contained in the seeds will be oxidized into small molecular substances such as hexenal, alcohol and ketone under the action of lipoxygenase, which is the soybean The main source of the beany smell. [0003] In order to breed new soybean varieties, select suitable parents for hybridization, and the seeds that are harvested for successful hybridization ...
