Embodiment

[0026] 1. Knockout of fadA gene in Aspergillus flavus

[0027] Search the NCBI database (https: / / www.ncbi.nlm.nih.gov / gene / ), and use bioinformatics methods to identify a new gene with unknown function numbered AFLA_018340 (GeneID: 7911513) in the genome of Aspergillus flavus , the gene is homologous to the fadA gene of Aspergillus nidulans, so it is named fadA gene (SEQ ID NO.1), and the FadA protein has the amino acid sequence shown in SEQ ID NO.2. In order to study the function of the fadA gene in Aspergillus flavus in the growth, development and toxicity of Aspergillus flavus, the fadA gene knockout fusion fragment was constructed in vitro, and the 4116bp DNA homologous fragment in the Aspergillus flavus genome was used by homologous recombination The argB gene fragment of Aspergillus flavus was replaced to knock out the fadA gene in the genome of Aspergillus flavus. figure 1 A shows the principle of this gene knockout.

Abstract

The invention discloses application of aspergillus flavus pathogenic gene fadA in screening of drugs capable of preventing and treating aspergillus flavus pollution. The application of the aspergillusflavus pathogenic gene fadA in the screening of the drugs capable of preventing and treating the aspergillus flavus pollution comprises the following steps: constructing a test-group aspergillus flavus culture system and a control-group aspergillus flavus culture system, wherein a drug to be screened is added into the test-group aspergillus flavus culture system; and then, culturing test-group aspergillus flavus and control-group aspergillus flavus under the same conditions for a period of time, comparing expression levels of the gene fadA or protein FadA in the aspergillus flavus in a test group and a control group, and determining the drug to be screened as a potential drug for inhibiting pathogenicity of the aspergillus flavus if the expression level of the gene fadA or the protein FadA in the aspergillus flavus in the test group is lower than the expression level of the gene fadA or the protein FadA in the aspergillus flavus in the control group to a certain extent. According to the screening method in the application of the aspergillus flavus pathogenic gene fadA in the screening of the drugs capable of preventing and treating the aspergillus flavus pollution, the inhibitioneffect of the drug on the aspergillus flavus is only necessary to be tested at a transcriptional level or a protein level, and the content of the aspergillus flavus is not required to be tested, thereby having test cycle and time shorted as well as use of various organic chemical reagents decreased; and thus, pollution to the environment is reduced, and possible toxicity to an experimental operator is greatly reduced.

Description

technical field [0001] The invention relates to genetic engineering, in particular to the application of the pathogenic gene fadA of Aspergillus flavus in screening and preventing the pollution caused by Aspergillus flavus. Background technique [0002] Aspergillus flavus is a widespread fungus of the genus Aspergillus. As a common pathogen of plants and animals, it not only causes great harm to agricultural production, feed industry, and animal husbandry, but also poses a serious threat to human health. Aspergillus flavus can infect many important crops, such as peanuts, corn, cotton, etc., which can pollute crops before and after harvest, causing huge economic losses to agricultural production around the world. According to the statistics of the Food and Agriculture Organization of the United Nations, about 25% of the grains are polluted by fungi every year, the most important of which is Aspergillus flavus. my country is the hardest-hit area for aflatoxin pollution.
