Embodiment 1

[0084] Rat hyperuricemia model was established by adding 5% potassium oxycyanate (Chengdu Dashuo Experimental Animal Co., Ltd.) to common rat diet for 2 weeks (references Lu, J., et al., Mouse models for humanhyperuricaemia: a critical review.Nature reviews.Rheumatology,2019.15(7):p.413-426.), in the 2-week experimental cycle, before starting the experiment, first use streptomycin containing 2mg / mL and 1 mg / mL of ampicillin in sterile drinking water fed rats (initial body weight of 120g-150g, Chengdu Dashuo Experimental Animal Co., Ltd., divided into 6 groups. Except for the 1-day group, the initial number of each group was 6 7 rats in the 1-day group) for three days, and then fed with normal sterile water for one day, then 1 day / time, 3 days / time, 5 days / time or 7 days / time, including 2×10 10 cfu Escherichia coli Nissle 1917 was administered with normal saline (about 200 μL), and the hyperuricemia group was administered with 0.2 mL of normal saline at a frequency of 1 day / tim...

Embodiment 2

[0088] In the rat hyperuricemia model established by adding 5% potassium oxycyanate (Chengdu Dashuo Experimental Animal Co., Ltd.) to the common rat diet for 2 weeks, in the 2-week experimental period, before starting the experiment Rats were first fed with sterile drinking water containing 2mg / mL streptomycin and 1mg / mL ampicillin (the initial body weight was 120g-150g, Chengdu Dashuo Experimental Animal Co., Ltd., divided into 6 groups. Each group The initial number was 6) for three days, and then fed with ordinary sterile water for one day, the allopurinol group was given 1.2mg / 100g high-intensity allopurinol by gavage at a frequency of 3 days / time (equivalent to In the sum of the upper limit of the human dose of allopurinol for 3 days and one-time administration), the Escherichia coli Nissle 1917 group was carried out at a frequency of 3 days / time containing 2×10 10 cfu Escherichia coli Nissle 1917 was intragastrically administered with normal saline (about 200 μL), and th...

Abstract

The present invention relates to the field of biomedicine, in particular to uricase or a homologous enzyme thereof as well as application of the uricase or the homologous enzyme. Through experiments,it is found that Escherichia coli Nissle 1917, which contains a novel non-homologous isomeric FAD-dependent urate oxidase, can be administered in a hyperuricemia rat model at a frequency of 3-5 days, and can still has ab effect of reducing blood uric acid of the rat model. The Escherichia coli Nissle 1917 is used, the purpose of using non-existing drug principles and avoiding daily use to alleviate hyperuricemia is achieved. The results show that Escherichia coli Nissle 1917 can be used for treating patients with hyperuricemia and gout.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to uricase or its homologous enzyme and application thereof. Background technique [0002] Hyperuricemia refers to a state of elevated uric acid in the blood, which is defined as a serum uric acid greater than 6.8 or 7.0 mg / dL. Long-term hyperuricemia can lead to gout. In recent years, it has been found that asymptomatic hyperuricemia may increase the risk of many diseases, such as cardiovascular disease and metabolic syndrome. These potential health problems have caused a heavy social medical burden. Therefore, hyperuricemia has received more and more attention in recent years. People with mild hyperuricemia can generally reduce their uric acid levels by controlling diet, losing weight, and changing lifestyle. However, many people with hyperuricemia are difficult to meet the above requirements because the state does not cause any disease manifestations. In this case, whether to use d...
