Abstract

The invention provides a preparation method of microparticles for preventing novel coronavirus and an application, and relates to the field of vaccines. Specifically, the preparation method comprisesthe following steps: 1) constructing recombinant plasmids for overexpression of a spike (S) protein; 2) packaging the recombinant plasmids into lentiviral particles; 3) infecting fibroblasts with thelentiviral particles to obtain tool cells for expressing of S protein; 4) performing X-ray irradiation on the obtained tool cells and a culture solution, and collecting supernatant after radiotherapyto obtain a mixture of required microparticles and apoptotic tool cell debris; and 5) performing centrifuging, concentrating and purifying on the obtained mixture to obtain a vaccine. According to theinvention, the microparticles released by radiotherapy induction tool cells are used as a carrier, so that the antiviral immunity of an organism can be directly activated under the condition of overexpression of the novel coronavirus S protein, and a neutralizing antibody for resisting the novel coronavirus is generated. According to the invention, an organism can be induced to generate a high-level neutralizing antibody immune response, and besides, good biological safety is achieved.
