Embodiment 1

[0086] According to the above-mentioned technical solution, this example provides an application of a molecular marker for identifying branch types of peppers in assisted breeding of pepper branch types.

[0087] The forward primer sequence of the molecular marker is: 5'-GGTTGAAGTTAATGGTGGTG-3', the sequence is shown in SEQ ID NO.1; the reverse primer sequence is: 5'-CTGTCGTAAAAGGAATAGCC-3', the sequence is shown in SEQ ID NO.2.

[0088] The specific method of this application includes the following steps:

[0089] Step 1, extracting the DNA of the pepper sample to be tested;

[0090] The pepper samples to be tested were pepper seeds of lines CM334, AA3, R9, P70, Z1 and GS6.

[0091] The specific process of extracting the DNA of the pepper sample to be tested is:

[0092] Put the pepper samples to be tested into appropriate containers, slowly pour water at 50°C to 55°C, and stir while pouring, so that the seeds are heated evenly. After 15-20 minutes, the water temperature dr...

Embodiment 2

[0108] According to the above-mentioned technical solution, this example provides an application of a molecular marker for identifying branch types of peppers in assisted breeding of pepper branch types.

[0109] The molecular marker is the same as that of Example 1.

[0110] The specific method of this application includes the following steps:

[0111] Step 1, extracting the DNA of the pepper sample to be tested;

[0112] The pepper samples to be tested were two true leaf stage plant leaves of lines CM334, AA3, R9, P70, Z1 and GS6.

[0113] The specific process of extracting the DNA of the pepper sample to be tested is:

[0114] Two pepper leaves at the true leaf stage were picked and frozen in liquid nitrogen. The genomic DNA of the leaves was directly extracted by the CTAB method, and then the DNA concentration and purity of the samples were measured by a micro-spectrophotometer, and detected by agarose gel electrophoresis at a concentration of 1%. DNA quality: Dilute th...

Abstract

The invention provides a molecular marker for identifying capsicum annuum branching types. A forward primer sequence of the molecular marker is 5'-GGTTGAAGTTAATGGTGGTG-3', and a reverse primer sequence of the molecular marker is 5'-CTGTCGTAAAAGGAATAGCC-3'. The invention relates to an application of the molecular marker to capsicum annuum branching type assisted breeding. The specific method of theapplication comprises the following steps: step 1, extracting DNA of a capsicum annuum sample to be detected; 2, taking the DNA extracted in the step 1 as a template, and performing PCR amplificationaccording to the molecular marker; and 3, detecting a PCR product obtained in the step 2 by adopting agarose gel electrophoresis with the mass percentage of 1% to obtain the length of a strip, and judging the result of the capsicum annuum branching type according to the length of the strip. According to the invention, genotype identification is performed on single plants and different capsicum annuum varieties in F2 population by using the marker, and the coincidence rate reaches 100%. Besides, the marker is used for identifying the branching types of 24 capsicum annuum varieties or breedingstrains with different branching types, and the coincidence rate is 100%.

Description

technical field [0001] The invention belongs to the technical field of vegetable breeding, relates to capsicum, in particular to a molecular marker for identifying branch types of capsicum and its application. Background technique [0002] Pepper (Capsicum annuum L.) belongs to Solanaceae and is a vegetable crop with a long history. In recent years, the annual planting area of pepper in my country has been 1.5 million to 2 million ha 2 At present, it has accounted for 8%-10% of the total vegetable planting area in the country, and plays an important role in the entire vegetable production. Capsicum has many applications in our daily life. It is not only used in pharmaceuticals, natural pigments, cosmetics, defense repellants, providing a variety of vitamins, minerals and nutrients necessary for human health, but also used as ornamental plants. Plant architecture is an important agronomic trait, and each plant has unique appearance and structural features that allow us to...
