Embodiment 1

[0030] Embodiment 1, the preparation of α-lactalbumin hydrolyzate

[0031] α-lactalbumin was dissolved in deionized water to prepare a 40mg / mL solution, adjusted to pH 2.0 with 5M HCl, added pepsin (250U / mg), and the ratio of enzyme to substrate was 1:30 (w / w) , under the conditions of 37°C and 170rpm for 2h; then adjust the pH of the system to 7.0 with 6M NaOH solution, add trypsin (8×USP), and the ratio of enzyme to substrate is 1:25 (w / w) , hydrolyzed for 4 hours at a temperature of 37° C. and a shaker speed of 170 rpm. After the hydrolysis, the pH of the system was adjusted to 7.0, and inactivated in a water bath at 95°C for 10 minutes. Then hydrolyzate is centrifuged 10min under the rotating speed of 12000rpm, gets supernatant and is complete hydrolyzate. Afterwards, carry out ultrafiltration with a filter diameter of 3kDa ultrafiltration tube (the specification is 50mL), and the filtrate is the hydrolyzate component <3kDa, which is freeze-dried to obtain a powder sam...

Embodiment 2

[0032] Example 2, mass spectrometric identification of VGINYW

[0033] The lyophilized <3kDa hydrolyzate fraction was redissolved with 0.1% formic acid solution, desalted with ZipTip desalting column, and the desalting column was equilibrated with 0.1% formic acid and acetonitrile to absorb the sample, washed with 0.1% formic acid, and then washed with 20% formic acid The sample was eluted with acetonitrile and 0.1% formic acid elution buffer, and the eluted sample was dried with a centrifugal concentrator. The dried sample was dissolved with 30 μL of loading buffer containing 2% acetonitrile and 0.1% formic acid, and analyzed by LC-MS.

[0034] NanoLC-MS conditions and process: The liquid chromatography analysis column is a C18 reverse-phase analysis column (75mm×15 cm×3μm); the peptide enrichment column is a C18 reverse-phase column (75mm×2cm×3μm). Mobile phase A is a mixed solution of 99.9% water and 0.1% formic acid, and mobile phase B is a mixed solution of 99.9% aceto...

Embodiment 3

[0035] Embodiment 3, the determination of ACE activity in vitro

[0036] The complete α-lactalbumin hydrolyzate, 50 , and the results are shown in Table 1.

[0037] Table 1 IC of different samples 50

[0038]

[0039]

[0040] The relative molecular weight of VGINYW is 750.85, and the IC of VGINYW is converted to molar concentration 50 is 15.1 μM.

Abstract

The invention discloses an alpha-lactalbumin hydrolysate and peptide fragment with in-vivo blood pressure lowering effect, the alpha-lactalbumin hydrolysate is prepared by hydrolyzing alpha-lactalbumin by pepsin and pancreatin, and then performing ultrafiltration separation on the hydrolysate to obtain the alpha-lactalbumin hydrolysate with the molecular weight of<3kDa of hydrolysate component. The amino acid sequence of the antihypertensive peptide is VGINYW. The alpha-lactalbumin hydrolysate and the antihypertensive peptide can be used as ACE inhibitors and can be used for preparing antihypertensive drugs and antihypertensive health-care products.

Description

technical field [0001] The invention belongs to the technical field of food and medicine, and in particular relates to an α-lactalbumin hydrolyzate capable of lowering blood pressure, a preparation method thereof, and a highly active blood pressure lowering peptide therein. Background technique [0002] Hypertension is a common cardiovascular disease that can cause various complications such as the heart, brain, and kidneys, as well as induce stroke, atherosclerosis, and coronary heart disease. WHO reported that in 2019 there were more than 1.13 billion hypertensive patients worldwide, and the prevention and treatment of hypertension is a global problem. Some clinical antihypertensive drugs are mainly angiotensin converting enzyme (ACE) inhibitors (such as lisinopril), diuretics (such as hydrochlorothiazide), angiotensin II (Ang II) receptor blockers (such as Olmesartan medoxomil) and calcium antagonists (nifedipine). These drugs can effectively lower blood pressure, but t...
