Problems solved by technology

[0013] However, even when epidermal cells are proliferated by these methods, the connection between the cells cannot be carried out, so it is still extremely difficult to form a standard planar cell layer on the surface cell layer of the living body.
[0014] In the past, synthetic macromolecular substances such as polyethylene and natural macromolecular substances such as collagen have been developed as cell culture beds, and natural macromolecular substances are included in synthetic macromolecular substances, but they cannot be said to be suitable for the proliferation of epidermal cells. Expect newer cell culture beds

Embodiment 1

[0077] The raw silk of silkworm was treated with 0.1% Na 2 CO 3 The aqueous solution is boiled for 2 hours under the condition of a solvent ratio of 100 times, and after washing, drying is carried out for degumming.

[0078] Dissolve the spun silk from which sericin has been substantially removed (the content of silk protein is more than 97%) in a solution with a molar ratio of calcium chloride, ethanol, and water of 1:2:8, and then dialyze it in water And desalination, make the aqueous solution of silk protein.

[0079] This aqueous solution was placed in a container for cell culture (made of polystyrene; flat bottom, 35 mm in diameter), and dried at room temperature to form a silk protein film (film-like) on the bottom of the container.

[0080] Next, the cell culture container on which the silk protein film was formed on the bottom was placed at room temperature and a relative humidity of about 90%, thereby adjusting the water content of the silk protein film to about 40%...

Embodiment 2

[0095] In this example, before the control, the wound dressing material was compared with silk protein β-type crystalline powder.

[0096] The α-type and β-type crystalline powders of silk protein were prepared as follows.

[0097] First, cast the silk protein aqueous solution obtained in Example 1 on an acrylic plate and air-dry to form an amorphous film with a thickness of about 30 μm.

[0098] Subsequently, the α-form and β-form were crystallized as follows.

[0099] In the crystallization, gelatinization is placed in a room with room temperature and a relative humidity of about 90% for one day so that the water content of the silk protein film is about 40%.

[0100] β-formation was carried out by spraying 70% ethanol aqueous solution on the silk protein coat.

[0101] These α-type and β-type fibroin coatings are dried, pulverized by physical means, and respectively made into an average particle size of 50 μm or less.

[0102] In order to understand the healing effects o...

Abstract

A novel epidermal cell growth activating material capable of not only permitting human and mammalian epidermal cells to be efficiently adhered to and grown on the surface of a baseplate but also promoting the adhesion between the cells themselves so as to form a planar layer. This material comprises a material containing silk fibroin of alpha type or one exhibiting an X-ray diffraction pattern of alpha -helix. It is remarkably excellent in the fundamental requirements for epidermal cell activation, such as cell adherence rate and growth, adhesion between cells themselves, and extensibility of cell layer.

Description

technical field [0001] The present invention relates to activating materials for promoting attachment, proliferation and cell layer formation of human and mammalian epidermal cells. [0002] The activated material can be used as a culture bed for epidermal cells, and as a defective skin repair material for living tissue or a wound covering material. Background technique [0003] Cells, which are the basic units of life, have superficial or latent self-proliferating capabilities. In multicellular organisms, each cell constituting the organism differentiates into cells responsible for various functions, and each expresses a predetermined growth pattern. [0004] Among them, the organism is made to form cells in the epidermis which is protected from external environmental changes so as to cover the dermis which is the lower layer of the epidermis in a layered form, and the cells have the ability to proliferate only by attaching to other organism tissues or substances that can r...
