Embodiment Construction

[0075] Example 1, nanometer gold and silver magnification and visual colorimetry detection of human hepatitis B and C virus pathogenic genes, the process is as follows:

[0076] (1) Prepare 0.2% PDITC (1,4-phenylene diisothiocyanate) solution

[0077] ①Mixed solvent

[0078] Pyridine: DMF (N, N-dimethylformamide) = 1:9 (note that they are all anhydrous solvents), for example: 0.19g PDC + 10ml pyridine + 90ml DMF is 100ml 0.2% PDITC solution.

[0079] ② Handling of slides

[0080] Wash the slides with concentrated nitric acid for 10 minutes, wash them with distilled water, and dry them in an oven.

[0081] ③ Preparation of oligonucleotide solution

[0082] Dissolve 5'-amino oligonucleotides with an optical density value of 1.0 (about 33ug) in 100ulTE at a concentration of about 3.35×10-5mol / L. 5'-amino oligonucleotide sequence: 5'-NH 2 -GGACTT CTC TCA ATT TTC TAG GG-3'

[0083] (2) Silanization of glass slides to capture the cross-linking of DNA and glass slides

Abstract

The present invention relates to kind of gene test technology, and is especially nanometer particle mark gene probe and its preparation and application. The nanometer particle mark gene probe is usedto replace traditional mark probe for practical application. The preparation of the nanometer particle mark gene probe incldues: the preparation of colloid gold or nanometer particle; the design, synthesis and modification of DNA probe molecule; and the self-assembling synthesis and enclosing of gold mark probe. The probe is suitable for use in various gene hybrid test, nanometer amplification and visual color development, and is especially suitable for application in low integration level diagnosis gene chip.

Description

1. Technical field [0001] The invention relates to a gene detection technology, more specifically, relates to a nanoparticle-labeled gene probe, a preparation method thereof and an application in gene hybridization. 2. Background technology [0002] Gene research led to the rapid development of the entire life science in the 20th century; in the 21st century, gene research will still promote the in-depth development of life science research. Genetic testing is not only crucial to biological research, but also of great significance to fields such as clinical medicine, pharmacy, agricultural technology, environmental monitoring, and legal identification. [0003] Genetic testing generally uses direct sequencing or hybridization testing. Direct sequencing is to directly separate, amplify, and purify gene fragments, and then use the principle of terminal chain termination reaction to determine the composition and sequence of each nucleotide in the nucleic acid. Sequencing meth...
