[0030] Various culture media were prepared in advance as required. Select growing way to be better, and come from the rubber tree variety (such as rubber tree clone Reyan 87-6-62) of good character, the radicle of the test-tube plantlet that obtains through the rubber tree anther culture embryoid body generation pathway, diameter is 1mm, in the absence of Cut into root segments with a length of 1.2 cm under bacterial conditions.

[0031] The root segment was inoculated in the callus induction medium, and cultured in the dark for 25 days at a culture temperature of 26±1° C. to induce the callus. The callus induction medium is an improved MS medium, and the content of anhydrous calcium chloride, potassium dihydrogen phosphate, manganese sulfate tetrahydrate, and magnesium sulfate heptahydrate in the MS medium is adjusted to anhydrous calcium chloride 250mg / L, potassium dihydrogen phosphate 400mg / L, manganese sulfate tetrahydrate 35mg / L, magnesium sulfate heptahydrate 500mg / L, a...

Abstract

The invention provides a method for regenerating plants through rubber-tree test-tube plantlet aseptic roots and somatic embryogenesis. The method mainly comprises the following steps: a rubber-tree test-tube plantlet aseptic radicle with the diameter between 0.8 and 1.2 millimeters is selected, cut into root segments with the length between 1.0 and 1.5 centimeters, subjected to dark culture in an embryogenic callus induction culture medium for 25 to 40 days, shifted into a subculture medium and subjected to dark culture for 15 to 30 days, transferred into a somatic embryo induction culture medium and subjected to dark culture for 20 to 50 days, transferred to a seedling-emergence culture medium and subjected to light culture for 20 to 50 days at a culture temperature between 25 and 28 DEG C, so as to grow into a complete plant. The method for regenerating plants through rubber-tree test-tube plantlet aseptic roots and somatic embryogenesis which is provided by the invention has the advantages that the method is simple in process flow, can adopt the roots of an excellent rubber tree variety as explants, induces plant regeneration via somatic embryogenesis, and establishes an excellent rootstock in vitro culture asexual reproduction system.

Description

technical field [0001] The invention relates to a plant tissue culture method, in particular to a method for obtaining regenerated plants through somatic embryogenesis by using the aseptic mycorrhizal roots of test-tube seedlings of rubber trees as explants. Background technique [0002] Rubber trees are perennial, cross-pollinated trees cultivated in tropical regions. Natural rubber, the main product of the rubber tree, is an important industrial raw material and strategic material, and occupies an important position in the national economy. Due to the limited land resources for planting rubber trees in tropical areas of our country, natural rubber is an irreplaceable scarce resource. Therefore, the breeding and promotion of high-yield rubber trees is the key to increasing the yield per unit area and ensuring the gradual increase of my country's total natural rubber production. [0003] With the development of plant tissue culture technology, since the 1970s, rubber tree ...
