Problems solved by technology

In recent years, Chinese scientific researchers have done a lot of work on the detection of tartrazine, but they are mainly concentrated on physical and chemical detection. However, these methods not only require expensive instruments and equipment, professional operators, but also have relatively high requirements for inspection materials. , and further sample preprocessing is required to perform
This can no longer meet the requirements of modern detection for fast, convenient and accurate
In recent years, research on pigment immunoassay methods has been carried out at home and abroad, but there are relatively few enzyme-linked immunoassay methods for tartrazine at home and abroad. In order to better detect tartrazine, it is necessary to prepare and develop a method that can target Tartrazine artificial antigen with higher sensitivity and better specificity antibody

[0031] (1) Preparation of tartrazine derivatives

[0032] Using succinic acid monoethyl chloride and 2,2-dimethyl-1,2-dioxane-4,6-dione as raw materials, after 4 steps of chemical reaction, tartrazine derivative TAR was prepared. The chemical synthesis steps are as follows:

[0033] Synthesis of diethyl 3-ketoadipate:

[0034]Take succinic acid monoethyl chloride (11.4g, 69.59mmol) and dissolve it in dichloromethane solution, and then add it dropwise within 1h to the stirring 2, 2-Dimethyl-1,2-dioxane-4,6-dione (8.98g, 62.33mmol) in the solution of Meldrum's acid, and then continue to stir for 1h. The reaction mixture was purified and concentrated to obtain diethyl 3-ketoadipate (13.4 g, 88%).

[0035] Synthesis of ethyl 1-phenyl-5-pyrazolone-3-propionate:

[0036] Diethyl 3-ketoadipate (13.4 g, 61.98 mmol) was dissolved in 200 ml of pure ethanol solution, and then phenylhydrazine hydrochloride (10.755 g, 74.38 mmol) was added to the solution in batches at room temp...

Abstract

The invention relates to a preparation method of an artificial antigen applied to tartrazine, belonging to the technical field of biochemical engineering. According to the invention, a derivative TAR of the tartrazine is prepared by taking ethyl succinyl chloride and 2, 2-dimethyl-1, 2-dioxane-4,6-dione as raw materials through four-step chemical reaction, and the TAR which contains no sulfonic acid group is used as a hapten and coupled with the carrier protein BSA or OVA by adopting an active ester method to prepare the artificial antigen for the tartrazine; the coupling rate of the conjugate is measured by adopting the spectrophotometry; the specific antibody which corresponds to the tartrazine is generated in an immunizing manner; and the enzyme-linked immunodetection method is established. The method successfully synthesized the artificial antigen of the tartrazine which can be fully used for immunoassay with concise and effective steps, thus providing necessary artificial antigen of the tartrazine for the further research of people, and meeting the domestic demand for the research of the tartrazine.

Description

technical field [0001] The invention discloses a method for preparing an artificial antigen suitable for tartrazine, which belongs to the technical field of biochemical engineering. Background technique [0002] Lemon yellow, English name: Tartrazine, Acid Yellow 23; chemical name is also tartrazine, hydrazine yellow, C.I. acid yellow 23; CAS number: 1934-21-0; molecular formula is C 16 h 9 N 4 Na 3 o 9 S 2 . Lemon yellow pigment (Tar) is one of the six food synthetic pigments approved for use in my country at present. Its molecular structure is an azo compound, and the products metabolized in the body are potentially harmful to the human body—aromatic compounds. Therefore, my country has strict restrictions on adding tartrazine to food. All meat and its processed products, fish and its processed products, etc. cannot use artificial synthetic pigments. Even within the applicable variety and scope, the dosage is strictly regulated, and excessive use is never allowed. ...
