Problems solved by technology

[0004] In view of the low content of baicalein and wogonin, which play a major role in curative effect in Scutellaria baicalensis, the content is about 1.7% and 0.6% respectively, it is difficult to separate and purify, and the existing hydrolysis transformation technology is complicated, the cost is too high, and the environmental pollution is serious, etc. Problem, the purpose of this invention is to provide a simple, safe, economical, efficient, endogenous enzymatic hydrolysis of baicalin and wogonin in Scutellaria baicalensis to prepare and separate and purify baicalein and wogonin

Embodiment 1

[0022] Precisely weigh 10 g of the traditional Chinese medicine Scutellaria baicalensis, add distilled water with a pH of 5, transfer it to a constant temperature negative pressure cavitation reactor, and enzymatically hydrolyze it at 35°C for 60 min with a vacuum of -0.04Mpa. After endogenous enzyme-induced transformation, the filtrate was filtered off, and 15 times the volume of 70% ethanol solution was added for negative pressure cavitation extraction at a vacuum degree of -0.06Mpa, and room temperature extraction was performed 3 times, 50 min each time. The extracts were combined and concentrated to dryness under reduced pressure. The conversion rates of baicalin and wogonin in the obtained crude extract were detected by HPLC to be 95.21% and 93.57%, respectively, and the contents of baicalein and wogonin could reach 9.65% and 2.10%. Compared with the original 1.79% and 0.61%, it has increased by 4.38 times and 2.46 times respectively.

Embodiment 2

[0025] Precisely weigh 10 g of the traditional Chinese medicine Scutellaria baicalensis, add distilled water with a pH of 5, transfer it to a constant temperature negative pressure cavitation reactor, and enzymolyze it at 50°C for 90 minutes, with a vacuum of -0.06Mpa. After endogenous enzyme-induced transformation, the filtrate was filtered off, and 20 times the volume of 70% ethanol solution was added for negative pressure cavitation extraction at a vacuum degree of -0.08Mpa, and room temperature extraction was performed three times, each time for 90 min. The extracts were combined and concentrated to dryness under reduced pressure. The conversion rates of baicalin and wogonin in the obtained crude extract were detected by HPLC to be 98.39% and 98.16%, respectively, and the contents of baicalein and wogonin could reach 9.83% and 2.33%. Compared with the original 1.79% and 0.61%, it is increased by 4.47 times and 2.85 times respectively.

Abstract

The invention relates to a method of preparing, separating and purifying baicalein and wogonin by endogenous enzymatic hydrolysis of baicalin and wogonoside in scutellaria, and aims at providing a simple, safe, economical and high-efficient method of extracting, separating and purifying the high-purity baicalein and wogonin. An adopted technical scheme is that the traditional Chinese medicine scutellaria is used as a raw material, a unique endogenous enzyme induction bioconversion technology, a negative-pressure cavitation extraction technology, a liquid-liquid extraction technique, a normal-phase silica-gel medium-and-low-pressure preparation liquid chromatography technology, a low-temperature crystallization and recrystallization technology and other high-efficient conversion, extraction, separation and purification technologies are used, and finally the high-purity baicalein and wogonin are obtained, wherein conversion rates of the baicalin and the wogonoside can respectively reach 98.39 % and 98.16 %, contents of the target products: baicalein and wogonin are increased by 4.47 times and 2.85 times, and a purity can be more than 98 %. The method overcomes disadvantages of large damaging effects on the baicalin and the wogonoside, low conversion rate and serious pollution existing in prior art, has advantages of simple production process, safe and environmental protection, high conversion rate and high target compound purity, and has a very important significance for development of medicines and health care products, and industrial production and application.

Description

technical field [0001] The invention relates to a method for preparing, separating and purifying baicalin and wogonin by hydrolyzing baicalin and wogonin in scutellaria baicalensis with endogenous enzymes, and belongs to the field of separation and purification of herbal medicines. Background technique [0002] Scutellaria baicalensis is the dried root of Scutellaria baicalens is Georgi, a plant in the Labiatae family. It is a precious Chinese medicinal material. It was first recorded in the first herbal book "Shen Nong's Herbal Classic" in ancient times. It has a long history of medicinal use and is widely used in my country. One of the traditional Chinese medicinal materials. It is cold in nature and bitter in taste, and belongs to the lung, gallbladder, stomach, and large intestine meridians. It has the effects of clearing away heat and dampness, stopping bleeding and preventing miscarriage, purging fire and detoxification, and lowering blood pressure. It can be used to tr...
