Problems solved by technology

Although the breeding quantity of raccoon dogs in our country is very large, the breeding method is still adopted at present, which requires a large number of breeding male raccoon dogs, which increases the feeding cost and wastes manpower and material resources. Moreover, the male raccoon dogs come into estrus earlier than the female raccoon dogs, and a large number of male raccoon dogs often appear in the later stage of mating. If male raccoon dogs cannot be used, raccoon dog artificial insemination technology can solve these problems
[0003] Due to the limitation of short domestication time, small breeding scale and species-specific differences, there are few domestic and foreign research materials on the collection of raccoon dog semen and related aspects.
At present, the artificial insemination of animals such as foxes and dogs is relatively mature. The research and promotion of artificial insemination of raccoon dogs is affected by the problem of semen collection.
At present, the research on the preservation of raccoon semen is mainly focused on the method of semen collection. There are few studies on the special diluent for the storage of raccoon semen at room temperature, and it has not been found in the CNKI

Embodiment Construction

[0008] 1. Preparation of diluent: Take 37.538g of glucose, 1.255g of sodium bicarbonate, 1.262g of ethylenediaminetetraacetic acid, 6.035g of sodium citrate, 0.758g of potassium chloride, 1.052g of bovine serum albumin, and 1.052g of N-acetyl-L - 0.169g of cysteine, 0.625g of penicillin, 0.505g of streptomycin, 0.150g of lincomycin, and 0.305g of spectinomycin were dissolved in three-distilled water, and the volume was adjusted to 1000ml volumetric flask, and filtered with a 0.22μm filter Use in time or subpackage in sterile bottles and freeze at -20°C.

[0009] 2. Semen collection: Fix the female raccoon dog in estrus on the semen collection frame, put her neck in the clasp and lock it. Let the male raccoon dog who picks the essence climb the female raccoon dog, and the male raccoon dog in Baoding is the assistant. Wash the vulva of the male raccoon dog with warm soapy water, rinse it with warm water, and then soak the hair on the lower abdomen of the male raccoon dog with a...

Abstract

The invention discloses a dilute solution used for preserving racoon dog seminal fluid at normal temperature, and a semen collection method. The dilute solution comprises following ingredients: 36 to 38g of glucose, 1.20 to 1.30g of sodium bicarbonate, 1.20 to 1.30g of ethylene diamine tetraacetic acid, 5.50 to 6.30g of sodium citrate, 0.65 to 0.85g of potassium chloride, 0.50 to 3.05g of bovine serum albumin, 0.05 to 0.30g of acetylcysteine, 0.50 to 1.20g of penicillin, 0.30 to 1.00g of streptomycin, 0.05 to 1.50g of lincomycin and 0.01 to 0.05g of spectinomycin; and a mixture of the ingredients above is diluted to 1000ml by adding triple-distilled water so as to obtain the dilute solution. Key steps of the semen collection method are that: female racoon dogs in heat are used as dummy teaser; conditioned reflex of male racoon dogs to artificial semen collection is formed by training; the training is repeated for 3 to 4 times until conditioned reflex is formed; and then the female racoon dogs are replaced by a female racoon dog-simulated dummy teaser which is filled with urine of female racoon dogs in heat so as to collect male racoon dog semen. The dilute solution and the semen collection method can be used for collecting racoon dog semen, diluting for preservation, realizing artificial insemination and obtaining offspring; and are capable of promoting the popularization of racoon dog artificial insemination technologies, and increasing economic benefits of racoon dog industry.

Description

technical field [0001] The invention relates to the technical field of reproductive biology, in particular to a diluent for storing raccoon semen at room temperature and a method for collecting semen. Background technique [0002] Raccoon raccoon is a precious fur animal. It has become one of the three major fur animals in my country due to its low feeding cost, resistance to rough feeding, and strong disease resistance. If male raccoon dogs cannot be used, raccoon dog artificial insemination technology can solve these problems. [0003] Due to the...
