Multi-channel optical imaging laser ionization source

An ionization source, optical imaging technology, applied in the direction of ion source/gun, material analysis by electromagnetic means, parts of particle separator tube, etc., can solve the problem that the sensitivity or spatial resolution is far less than MALDIIMS, and cannot be separated and analyzed. It can be used in conjunction with the mass analyzer and other problems, so as to achieve the effect of widening the range of use, good sensitivity and resolution, and easy ionization.

Problems solved by technology

[0004] The MALDI ionization source device is an ionization source under a vacuum system. It has high requirements on the instrument and device. It cannot be connected to various mass analyzers quickly and conveniently, and it cannot be used in conjunction with other separation and analysis methods.
Although the current atmospheric ionization source technology desorption electrospray ionization (DESI) can directly mass spectrometer imaging under atmospheric conditions without sample pretreatment, it is far inferior to MALDI IMS in terms of sensitivity and spatial resolution.

Embodiment Construction

[0034] The present invention will be further described below in conjunction with the accompanying drawings, but the present invention is not limited to the following embodiments.

[0035] Such as figure 1 As shown, the multi-channel optical imaging laser ionization source provided by the present invention includes an ultraviolet pulsed laser source 1 , a conductive glass substrate 4 and a microscope 7 . The conductive glass substrate 4 is arranged on the optical path of the microscope 7, and is irradiated on the upper part of the conductive glass substrate 4, and the optical information of the sample can be observed through the eyepiece 8. The conductive glass substrate 4 is arranged on a three-dimensional mobile platform (not marked in the figure), In order to facilitate the position control of the conductive glass substrate, in the present invention, the conductive glass substrate 4 is arranged parallel to the ion transfer tube 5 . The laser light emitted by the ultraviolet...

Abstract

The invention discloses a multi-channel optical imaging laser ionization source. The ionization source comprises a light source, a light permeating substrate and an optical detection system. The light permeating substrate is arranged on a light path of the optical detection system. A laser emitted by the light source is reflected by a reflecting mirror to form reflected light, and the reflected light is focused through a focus objective lens and then irradiates the light permeating substrate. Under the premises that sampling time is not prolonged and the character of a sample is changed, the multi-channel optical imaging laser ionization source can be used in a vacuum environment and meanwhile can carry out ionization under the atmosphere or sub-vacuum condition, and accordingly the using range of the ionization source is enlarged. By means of a matrix, the sample can be ionized more easily through the excitation of the light source, and accordingly better sensitivity and a better resolution ratio can be obtained. Due to the fact that the light source irradiates from the back face of the substrate, the sample can be ionized by utilizing a small laser spot, and therefore the method has the ultrahigh space resolution ratio.

Description

technical field [0001] The invention relates to a multi-channel optical imaging laser ionization source. Background technique [0002] As a new in situ analysis technique, mass spectrometry imaging is widely used in the analysis of "structure, space and time distribution" of molecules in cells or tissue slices of biological samples. To put it simply, this technology uses mass spectrometry and is equipped with special mass spectrometry imaging software to directly analyze the sample to obtain a two-dimensional ion density map of any compound with a specified mass-to-charge ratio (m / z). The composition, relative abundance and distribution of pharmaceutical compounds, lipids and proteins, etc.) have been comprehensively, rapidly and high-throughput multiple analysis, so as to obtain the spatial distribution of potential biomarkers, the distribution of candidate drugs in target tissues etc., which are of great significance to the discovery of biomarkers and the monitoring of co...
