Abstract

The method includes: selecting a TCA-acetone technique to extract Magnaporthe oryzae mycelium protein, rinsing precipitates by a DTT-containing precooling acetone solution, and using a 4-times volume precooling acetone solution to precipitate sample protein dissolved in a lysate solution, making use of acetone to perform secondary precipitation on the protein sample in a hydration solution, and increasing the dose of a TCA-acetone reagent and prolonging the treatment time to effectively remove impurities in the protein sample so as to obtain pure sample protein precipitates, and finally adding the lysate solution to prepare the high quality sample suitable for two-dimensional electrophoresis.
