Problems solved by technology

It is widely used in Europe, but so far in my country, phytase has not been widely used in feed. The main reasons are: 1. The content of phytase in natural materials is too low to be produced in large quantities. The production cost is expensive; 2. The stress resistance of phytase, especially the thermal stability, cannot fully meet the requirements of feed and feed processing

Embodiment Construction

[0025] A polyhedrin-encapsulated phytase of the present invention will be further described in detail below in conjunction with specific examples.

[0026] A polyhedrin-coated phytase is composed of a phytase aPPA molecule and a wrapping layer, the wrapping layer is an insectoid polyhedrin, and the phytase aPPA molecule is wrapped in the solid molecule In the insectoid polyhedrin protein; the polyhedrin-encapsulated phytase has a particle size of 0.1-1 μm, and the phytase protein concentration accounts for 10%-70%.

[0027] The polyhedrin-encapsulated phytase is made by the following method, and the specific steps are as follows:

[0028] A. Construction of synthetic aPPA gene

[0029] Utilizing the codon bias exhibited by the viral genome, the determination of the preferred codon can be determined by exhaustively investigating the sequence of the available virus or insect gene, such as investigating the 20 amino acids of the polyhedrin protein and the coding codons of the st...

Abstract

The invention discloses polyhedrin coated phytase. A cytoplasmic polyhedrin gene and a phytase gene are respectively constructed under PH and p10 promoters of a pFastBac Dual carrier simultaneously by utilizing a baculovirus expression system; an SP guide gene is introduced in the front of the phytase gene; the SP guide gene is recombined with a virus skeleton; and therefore, recombinant baculovirus is obtained. Required polyhedrin coated phytase particles can be directly generated in cells by using the single virus to infect Sf9 or Sf21 insect cells; furthermore, the recombinant virus can also be amplified; the polyhedrin coated phytase particles are 0.1-1 mu m in particle size; and the polyhedrin coated phytase particles have the characteristics of being acid-resistant, high-temperature-resistant, durable, slow to release and the like.

Description

technical field [0001] The invention belongs to the field of biotechnology and relates to a polyhedrin-encapsulated phytase. Specifically, the invention relates to the construction of a recombinant polyhedron baculovirus co-expressing a phytase gene and a polyhedrin gene and a polyhedron-wrapped phytase. Background technique [0002] Phytase is a new feed additive for monogastric animals that can hydrolyze phytate phosphorus in feed into inorganic phosphorus and inositol. It is mainly used in monogastric animals such as pigs, chickens, ducks and aquaculture animals. It has a wide range of applications. The demand is high. It can increase the utilization rate of phosphorus in plant feed by 60%, reduce the excretion of phosphorus in feces by 40%, and at the same time reduce the anti-nutritional effect of phytic acid. Therefore, adding phytase to feed is of great significance to improve the production efficiency of livestock and poultry industry and reduce the pollution of ph...
