Embodiment 1

[0085] A Legionella pneumophila type I detection kit is provided that contains the reagents described in Tables 1-3 above.

Embodiment 2

[0087] Legionella pneumophila type I detection method, the method comprises the following steps:

[0088] S21. Extraction of DNA from the sample to be tested:

[0089] a. Bacterial enrichment culture: the collected specimens are placed in a constant temperature room at 37°C for 3 hours of bacterial enrichment culture;

[0090] b. DNA lysis: Take 1ml of enrichment solution and add 50μL of DNA extraction solution, and repeatedly pump 3 times to fully mix the specimen;

[0091] According to the above steps a-b, extract the 4 samples to be tested as shown in Table 4 below.

[0092] S22. Real-time fluorescent PCR:

[0093] a. Sample group to be tested: provide the Legionella pneumophila type I detection kit in Example 1, and take 5 μL of the DNA sample extracted in step S21, add it to the PCR reaction solution in the kit, and test on the machine.

[0094] b. Negative control group: take 5 μl of the negative control solution of the Legionella pneumophila type I detection kit, add...

Abstract

The invention discloses a detection kit and a detection method for I-form legionella pneumophila. The detection kit comprises a PCR (polymerase chain reaction) reaction liquid, wherein the PCR reaction liquid comprises first reaction liquid and second reaction liquid which are separated from each other, and the second reaction liquid contains enzyme; the first reaction liquid contains a primer probe for carrying out fluorescent quantitation PCR on target genes of I-form legionella pneumophila. The detection method comprises the steps of extracting DNA of a to-be-detected sample, carrying out real-time fluorescent PCR and comparing and judging a fluorescence signal detection result with a control group. According to the detection kit, the first reaction liquid containing the primer probe of the I-form legionella pneumophila and the second reaction liquid containing enzyme can be directly mixed to carry out real-time fluorescent PCR, so that the I-form legionella pneumophila can be rapidly detected; furthermore, the pollution is avoided, and the sensitivity is high.

Description

technical field [0001] The invention belongs to the technical field of sanitation prevention virus detection, and in particular relates to a detection kit for Legionella pneumophila type I and a detection method for Legionella pneumophila type I. Background technique [0002] Legionnaires' disease is one of the newly discovered infectious diseases in the past 20 years, and the disease has become a public health problem of general concern in countries all over the world and in my country. Legionella pneumophila is the main pathogen that causes Legionnaires' disease in humans, and 14 serotypes have been found, among which Legionella pneumophila type Ⅰ is the most common infectious type. Legionella pneumophila (legionella pneumophila) is a short coccus bacillus of the genus Legionella, also known as Legionella. Legionella pneumophila can invade amoebas, or human macrophages. It mainly exists in warm and humid natural environments, and is also widely distributed in artificial ...
