Abstract

The invention discloses a preparation method of high-purity lycium polysaccharides. The method includes the following steps that lycium barbarums or lycium-barbarum waste residues are crushed, sievedand then subjected to subcritical degreasing, high-speed shearing, low-temperature wall breaking, water extraction, high-speed centrifugation, microfiltration-membrane separation, microfiltration-membrane concentration, inorganic/organic two-phase extraction, impurity removal with macroporous adsorption resin, ethanol precipitation and freeze-drying to obtain the high-purity lycium polysaccharides. It is tested that the active ingredient, namely, lycium barbarum polysaccharide, in the extract is not less than 80%. The lycium polysaccharides are separated by using a 30 kD microfiltration membrane, only the lycium polysaccharides with the molecular weight less than 30 kD are obtained, the lycium polysaccharides with the molecular weight not less than 30 kD are removed, as the lycium polysaccharides with the molecular weight not less than 30 kD are metabolized slowly in vivo, massive accumulation is resulted, 'internal heat' can be induced, and the biological activity of the lycium polysaccharides with the molecular weight not less than 30 kD is low. The four purification technologies are adopted, such as nanofiltration-membrane concentration, two-aqueous-phase extraction, impurity removal with macroporous adsorption resin and ethanol precipitation, and the purity and quality of the obtained lycium polysaccharides are ensured.

Description

technical field [0001] The invention relates to a preparation process of wolfberry glycopeptide, in particular to a preparation process of high-purity wolfberry glycopeptide, and belongs to the technical field of extraction and separation of active ingredients of traditional Chinese medicines. Background technique [0002] Lycium barbarum is a solanaceous plant Lycium barbarum ( Lycium barbarum L. ) dry ripe fruit, its name comes from "Shen Nong's Materia Medica", it has a history of more than 3000 years, and it is the earliest medicinal and edible substance approved by the Ministry of Health. There are many records of the medicinal effects of Lycium barbarum in traditional Chinese medicine classics. Modern pharmacology and natural product chemistry studies have shown that Lycium barbarum glycopeptide (LBP) is one of the main active components of Lycium barbarum. Glycopeptide structure, the content is 3%~9%. Studies have shown that wolfberry glycopeptide is a water-soluble...
