Embodiment 1

[0066] 1. Experimental materials and methods

[0067] 1.1 Materials

[0068] 1.1.1 Test plants and strains

[0069] The tobacco used for the test was Nicotiana benthamiana preserved in our laboratory. Nicotiana benthamiana was planted in the greenhouse at a temperature of about 24°C and a humidity of about 70%, with 14 hours of light and 10 hours of darkness. The tobacco was grown for 25 days before being used for the test. The peppers tested are laboratory inbred varieties. The pepper seeds were placed in a large petri dish covered with absorbent paper and placed in a constant temperature incubator at 25°C for about 7 days. After the pepper seeds germinated, they were planted in In the flower pot with the substrate, the film is used to keep moisture. The post-emergence management was the same as that of Nicotiana benthamiana.

[0070] The test strains Phytophthora capsici LT1534 and SD33 were preserved in our laboratory; the Agrobacterium strains of Bax and INF1 were kindl...

Embodiment 2

[0173] In this example, the function of RxLR129113 was verified by silencing acetaldehyde dehydrogenase (NbALDH).

[0174] 1. Transient silencing of NbALDH gene in Nicotiana benthamiana

[0175] In order to verify whether ALDH affects the function of RxLR129113, the capsicum ALDH gene was compared with Nicotiana benthamiana genomic DNA, and the ALDH with the highest homology rate in Nicotiana benthamiana was NbALDH (nucleotide sequence shown in SEQ ID No.45).

[0176] From the N-terminal and C-terminal of NbALDH, two specific sequences NbALDH-1 (nucleotide sequence as shown in SEQ ID No.46) and NbALDH-2 (nucleotide sequence as shown in SEQ ID No.47) and NbALDH-2 (nucleotide sequence as shown in SEQ ID No.47) were screened out. shown), and constructed into the pTRV2 virus silencing vector, and after the sequence verification was correct, it was transferred to Agrobacterium GV3101.

[0177] 2. Verification of functional changes of RxLR129113 after silencing acetaldehyde dehydro...

Abstract

The invention relates to the field of molecular biology, and specifically discloses a fungus effector, a coding gene and application thereof. Through experimental researches, it is found that the effector RxLR129113 (nucleotide sequence as shown in SEQ ID No.1) from Phytophthora capsici can cause the hypersensitive reaction (HR) and inhibit Bax-induced programmed cell death (PCD) and inhibit INF1-induced cell death on nicotiana benthamiana. According to the structure of RxLR129113, RxLR129113 is truncated, and four truncated mutants (RxLRW1, RxLRW1-2, RxLRW1-3 and RxLRW1-4) are subjected to functional verification; and it is found that RxLRW1-3 is a key functional domain that inhibits cell death caused by Bax and INF1. The nuclear output and nuclear input functions of RxLR129113 and RxLRW1-3 are verified, and the result shows that when RxLR129113 is located in cytoplasm, HR can be caused and the ability of inhibiting Bax and INF1 is strong, and when RxLRW1-3 is located in cytoplasm, the ability of inhibiting Bax and INF1 is strong.

Description

technical field [0001] The invention relates to the field of molecular biology, in particular to a fungus effect factor and its coding gene and application. Background technique [0002] Once the diseases caused by oomycetes occur, they cannot be controlled, resulting in the loss of large areas of crops and bringing huge economic losses to human beings. In recent years, the research on the whole genome of oomycetes has made great progress, but the research on effectors is still the focus. From the results of whole genome sequencing, it is found that the effector proteins secreted by oomycetes are relatively complex, and hundreds of RxLR effectors contribute their power in various ways in the pathogenic process. Studying the pathogenic mechanism of pathogenic bacteria is of great significance to the development of new fungicides and the effective prevention and treatment of oomycete diseases. [0003] When plant pathogens infect host plants, they will activate the plant's o...
