Problems solved by technology

[0003] The current method of treating mastitis is relatively single, and injecting antibiotics into the mammary gland is a common method
However, the abuse of antibiotics and the emergence of superbugs make the treatment of cow mastitis more difficult
Hydroxycamptothecin (HCPT) is a class of natural active ingredients extracted from plants. It has anti-inflammatory and anti-cancer effects. As a natural anti-inflammatory drug, it can be used to treat inflammatory diseases. However, due to its poor water solubility, Limit its promotion and use

Embodiment 1

[0028] Example 1. Determination of Zeta Potential and UV Absorption of GO / CS / Pep / Anti-TLR4 / HCPT

[0029] The GO / CS / Pep / Anti-TLR4 / HCPT prepared in the experimental step (3) was taken to measure Zeta potential and ultraviolet absorption. like figure 1 As shown in A, the Zeta potential of GO is -42.1±0.62 (mV), the Zeta potential of GO / CS after CS modification of GO is +45.0±0.75 (mV), and the Zeta potential of Pep and Anti-TLR4 after modification is +30.70 ±0.89mV, its Zeta potential becomes smaller on the basis of GO / CS. like figure 1 As shown in B, compared with GO, the UV absorption curve of GO / CS / Pep / Anti-TLR4 / HCPT showed characteristic peaks of Pep (450nm and 475nm) and characteristic peaks of HCPT (267nm).

Embodiment 2

[0030] Example 2. Determination of GO / CS / Pep / Anti-TLR4 drug-loading capacity and drug-loading efficiency

[0031] Carry out the experiment according to the above (4) steps. The LC and LE of HCPT in GO / CS / Pep / Anti-TLR4 / HCPT were calculated according to the formula, and the LC was 1.68±0.23mg / mg, and the LE was 94.31±0.30%.

Embodiment 3

[0032] Example 3. Determination of drug release under different pH conditions of GO / CS / Pep / Anti-TLR4 / HCPT

[0033] Carry out experiment according to above-mentioned (5) step, observe the release of medicine under different pH conditions at different times. The results are shown in Figure 3, GO / CS / Pep / Anti-TLR4 / HCPT in the PBS buffer solution of pH=7.4 and pH=8.0, the cumulative release rate of HCPT at 24h was 20.75% and 10.94%, respectively, at pH=5.8 In the PBS buffer solution, the cumulative release rate of HCPT was 65.47% at 24 hours.

Abstract

The invention belongs to the field of nano drugs, and relates to preparation and application of a graphene oxide-loaded hydroxycamptothecine nano targeting drug. Graphene oxide is mainly adopted as acarrier, and the nano targeting drug is prepared through modification of targeting molecules and loading of the drug. In an experiment, the preparation comprises the steps: performing covalent modification on graphene oxide (GO) through fluorescein-labeled Caspase-3-specific substrate peptide (Pep), chitosan (CS) and Toll-like receptor 4 antibody protein (Anti-TLR4), loading hydroxycamptothecine (HCPT), and preparing the nano targeting drug (GO / CS / Pep / Anti-TLR4 / HCPT) through adoption of GO as the carrier. The drug loading and drug releasing properties of the drug are judged through detection of the drug loading capacity, drug loading efficiency of GO / CS / Pep / Anti-TLR4 / HCPT and release of the drug under different pH conditions, the drug targeting property of GO / CS / Pep / Anti-TLR4 / HCPT is verified through a laser confocal experiment, and the curative effect of GO / CS / Pep / Anti-TLR4 / HCPT is observed through detection of the content of inflammatory factors and the activity of related enzymes. The nano targeting drug has good stability, and can be applied to treatment of bovine mastitis.

Description

technical field [0001] The invention relates to the preparation and application of a graphene oxide-loaded hydroxycamptothecin nano-targeted drug, belonging to the field of nano-medicines. Background technique [0002] Cow mastitis is mostly a syndrome caused by pathogenic microbial infection, mechanical damage, physical and chemical changes, and physiological and metabolic disorders in mammary gland tissue, including mammary gland infection and inflammation, microcirculation and immune dysfunction. Dairy cow mastitis is a complex multi-pathogenic disease that affects the production and feeding of dairy cows and the quality of dairy products worldwide. It has a high incidence and causes significant economic losses to the dairy cow production industry and related food industries. According to statistics, mastitis in dairy cows causes losses of up to tens of billions of dollars every year worldwide. [0003] The current method of treating mastitis is relatively simple, and in...
