Assays and reagents for the detection of soluble gp120

PendingUS20260251654A1Blood plasmaReceptor binding site
Soluble gp120 (sgp120) is associated to HIV-1-induced immune dysfunction, such as chronic immune activation. However, the detection of sgp120 in biological fluids such as plasma, serum, cervicovaginal fluids, and cerebrospinal fluids is challenging. The present application relates to novel methods and kits for the detection of sgp120 in biological fluids. These methods and kits are based on the use of specific combinations of sgp120-binding molecules recognizing particular domains of sgp120, notably the N-terminal 8-stranded β-sandwich structure, the constant region 1 and 2 (C1-C2) portion, the bridging sheet of the Co-Receptor Binding Site (CoRBS), and / or the CD4-binding domain.

Novel latilactobacillus curvatus strain having immunoenhancing efficacy and immunoenhancing composition comprising same

The present invention relates to a novel Latilactobacillus curvatus strain WiKim0169 having immunoenhancing efficacy, and an immunoenhancing composition comprising same. In the present invention, it was found that the Latilactobacillus curvatus strain WiKim0169, which is a novel kimchi-derived strain, significantly increases splenic NK cell activity and increases the production of inflammatory mediators, immunoglobulins, and cytokines, thereby alleviating immunosuppressive responses. In addition, it was confirmed that the Latilactobacillus curvatus strain WiKim0169 has an effect of alleviating immune dysfunction by increasing the mRNA expression levels of iNOS, COX2, TNF-α, IL-1β, IL-6, IL-2, IL-12, and IL-10 in splenic tissue and increasing the phosphorylation levels of NF-κB and IκBα. Therefore, according to the present invention, the composition comprising the Latilactobacillus curvatus strain WiKim0169, which is a novel kimchi-derived strain, as an active ingredient is expected to be usefully employed as a pharmaceutical and functional food composition for immunoenhancement.

Microneedle patch for inducing immune tolerance to treat rheumatoid arthritis

The application discloses a polymer microneedle patch co-loaded with self-antigen peptides and immunomodulators and a preparation method thereof, and studies the ability of the polymer microneedle patch to reverse immune dysfunction of rheumatoid arthritis (RA). The preparation method is as follows: a biocompatible polymer matrix material and a self-antigen are dissolved in pure water, and then an immunomodulator is added to obtain a suspension by stirring. The suspension is added to a mold to form needle tips, a backing layer solution is poured after drying, and finally the microneedle patch is demolded. Animal experiments show that the microneedle has sufficient mechanical strength to deliver drugs transdermally, effectively induces tolerogenic dendritic cells at the drug delivery site, further activates the differentiation of regulatory T cells (Treg), significantly up-regulates anti-inflammatory cytokines, down-regulates pro-inflammatory cells, antibodies and cytokines, effectively restores the immune balance of rheumatoid arthritis, and finally almost completely eliminates the symptoms and inflammatory infiltrates of rheumatoid arthritis. The microneedle preparation method is simple and rapid, and has a good clinical transformation prospect.

Biomarkers, kits and uses thereof for assessing immune dysfunction in hiv-infected individuals

This invention relates to the biomedical field, specifically to biomarkers, kits, and their applications for assessing immune dysfunction in HIV-infected individuals. This invention discloses the application of the molecular marker SMPD1 and / or substances that detect said molecular marker in the assessment, auxiliary assessment, screening, and risk stratification of immune dysfunction in HIV-infected patients with hyperlipidemia. This invention is the first to identify an immunometabolic paralysis subtype in HIV-infected individuals with hyperlipidemia. SMPD1 expression was significantly upregulated in these subtype patients, and its high expression was significantly associated with the risk of immune reconstitution failure. A combined diagnostic model based on SMPD1 and triglycerides demonstrated excellent diagnostic efficacy. Therefore, substances that detect SMPD1 can be used for the assessment and risk stratification of immune dysfunction in HIV-infected patients with hyperlipidemia. This invention provides an effective tool for the early identification and risk stratification of these patients and has broad clinical application prospects.

Activation-dependent immune dysfunction traits enable identification of prodromal Parkinson's

PendingUS20260146997A1Disease diagnosisBiological testingPeripheral blood mononuclear cellRapid eye movement sleep behaviour disorder
Idiopathic Parkinson's disease (iPD) is a multi-system disorder, and the debilitating motor stage of iPD can be preceded for years by a prodromal stage characterized by non-motor symptoms like REM sleep behavior disorder (RBD) and gastrointestinal symptoms. Widespread immune dysregulation has been reported in clinically diagnosed iPD, but the existence of immune deficits during the prodromal stage has yet to be thoroughly investigated. Here, it was shown that multiple stages of iPD, including the pre-motor prodromal stage, can be stratified according to the immuno-metabolic response to stimulation of peripheral blood immune cells ex vivo. Peripheral blood monocytes from RBD patients displayed increased stimulation-dependent secretion of inflammatory cytokines, including TNF, IL-1β, and IL-8, which peaks in the prodromal stage and successively diminishes as PD progresses. Furthermore, T lymphocyte mitochondrial health was correlated with stimulation-evoked cytokine secretion across patients with RBD, early-stage iPD, and moderate-stage iPD. The results disclosed here have broad implications for mechanistic understanding of how peripheral inflammation may drive disease progression, and it reveals novel biomarkers to enable patient stratification and progression monitoring for clinical trials based on immune endophenotypes.
