The method comprises the following steps of adding a sample reaction solution into the PCR reaction tube, and sealing the PCR reaction tube with a sealing film; inserting the optical fiber bundle end into the PCR reaction tube through the sealing film; and irradiating a photo-thermal heating column by laser with different energy, irradiating a sample solution by the light source excitation path, and collecting fluorescence generated by the sample solution by an optical sensor.
