Embodiment 1

[0027] This example proves that mLR12 (the murine polypeptide corresponding to LR12) can significantly relieve the edema symptoms of the rat tail model of lymphedema.

[0028] 1. Mouse tail lymphedema model and administration of LR12

[0029] The method for establishing the rat tail model of lymphedema: adult male (4-6 weeks old, C57) mice were anesthetized by intraperitoneal injection of 0.1 mL of 10% chloral hydrate, and the root of the tail was sterilized with 75% ethanol. The skin was incised at 1 cm above and the deep lymphatic structure of the tail was destroyed. For more specific modeling methods, please refer to the literature "Zampell JC, Yan A, ElhadadS, Avraham T, Weitman E, Mehrara BJ.CD4+Cells Regulate Fibrosis and Lymphangiogenesis in Response to Lymphatic Fluid Stasis.PLoS One.2012; 7:e49940" .

[0030] Grouping and administration method: After the mice recovered naturally for 2 weeks, the tail diameter of the mice at the incision 1 cm was measured, and then t...

Abstract

The invention provides application of a drug intervention target in preparation of a drug for treating and / or relieving lymphedema. The target is a TREM1 gene and an encoding protein thereof. The invention further provides application of the drug intervention target in preparation of a drug for treating tissue fibrosis and / or fat deposition and application of the drug intervention target in preparation of a drug for inhibiting expression of harmful inflammatory factors or reducing the number of pro-inflammatory macrophages. The targets are TREM1 genes and encoding proteins thereof. The drug is a polypeptide drug capable of interfering the binding of TREM1 and a ligand thereof, and on the basis, drugs, including polypeptides, designed aiming at the targets can be used for treating and relieving the lymphedema. The invention provides new drug targets and a new drug choice for treatment and / or alleviation of the lymphedema.

Description

technical field [0001] The invention belongs to the field of biomedicine, and specifically relates to a drug intervention target for treating and / or alleviating lymphedema and an application thereof. Background technique [0002] Lymphedema is the edema caused by the retention of lymphatic fluid in the tissue due to the obstruction of lymphatic return. According to the statistics of the World Health Organization, lymphedema ranks 11th among common chronic diseases, and ranks 2nd among disabling diseases. There are about 170 million patients with edema, and the incidence of patients with lymphedema in my country is still rising year by year. [0003] Lymphedema generally manifests as progressive swelling of subcutaneous adipose tissue of limbs, chronic inflammatory response of adipose tissue, pathological fibrosis of adipose tissue and excessive fat deposition and other pathological processes. Lymphedema often brings heavy physical and psychological burdens to cancer patients...
