Universal microarray system

The present invention provides a simple, cost-effective, universal method for determining and/or quantifying differences in nucleic acid levels between two or more test mixtures without prior knowledge of the sequence of the nucleic acids of interest. The method involves providing a universal microarray containing a plurality of spots, where each spot contains a plurality, or pool, of different oligonucleotide probes having at least three distinct portions: a universal sequence portion, a short central variable “wobble” sequence portion, and a unique sequence portion. A set of probes is synthesized such that the universal sequence portion is the same for every probe, and all possible permutations of the wobble sequence and unique sequence portions are represented in approximately equal concentrations in the set. The probes are pooled on the universal microarray such that probes in a given spot have the same unique portion and every permutation of the short wobble, while probes on different spots have different unique portions. Primers complementary to the universal and wobble portions of the probes are used to synthesize, for example, cDNA from an mRNA preparation. The cDNA is hybridized to the universal microarray, and through the use of differential labeling, is identified and/or quantified.

The invention discloses a method for determining the content of drug components in a mixture drug. The mixture drug is prepared from a first drug and a second drug. The preparation method comprises the following steps: (1) preparing a group of standard samples with different component contents; (2) collecting the terahertz time-domain spectrums of each standard sample and air, and respectively converting the terahertz time-domain spectrums into absorption coefficients and refractive indexes of corresponding frequency domain; (3) determining the terahertz band confidence interval of a test model; (4) selecting a group of optical parameters in the terahertz band confidence interval of each standard sample in the test model to be used as effective optical parameters and establishing a corresponding effective optical parameter matrix; (5) establishing a test model of the corresponding relationship between the content of the first drug and the effective optical parameters of each standard sample; and (6) inputting the effective optical parameter matrix of the sample of the to-be-tested mixture drug to the test model to obtain the content of the first drug in the to-be-tested mixture drug. By utilizing the method, the content of the drug components in the mixture drug can be quickly detected in a lossless way.

Method for detecting the presence or absence of pathogenic Staphylococci in a test sample, including test mixture with micro particles

A presence / absence test for Staphylococcus aureus (S. aureus) involves placing a first generation test sample in a solution that will clot in the presence of S. aureus. The solution contains components that will selectively grow S. aureus and also contains clotting factors that will react with S. aureus, if S. aureus is present in the sample, to clot the solution. Examples of specimen samples that can be tested include nasal swabs and lesion swabs, among others. The test can also be modified to detect the presence or absence of methicillin resistant S. Aureus (MRSA). The addition of micro particles having a size in the range of about 0.1 micron to about 1.0 mm provides localities where the bacteria agglomerate, thereby significantly decreasing the clotting time, and providing a significantly stronger clot. The micro particles can be used in other bacteria tests to accelerate the production of an end result. Such other tests can include a vancomycin-resistant enterococcus test; a Group B Streptococcus test; a test for hemolytic E. coli; and a test for Listeria monocytogenes, to name a few. These tests are all performed in a liquid broth-type reagent mixture and do not necessarily involve clotting of the broth.
