Embodiment Construction

[0007] Thaw the frozen North Pacific squid ink sac at room temperature, wash 4 times with clean water, remove the surface debris, squeeze the ink sac, and take the squid ink. Weigh 1Kg of squid ink, add 2Kg of distilled water to stir, wash, filter with coarse filter cloth to remove impurities such as ink glands and inner membrane of ink sac. Adjust the pH to 7.2 with NaOH aqueous solution, add 10 g of papain under stirring, and hydrolyze the enzyme in a water bath shaker at 60°C for 20 hours. The reaction was terminated by heating at 90-100°C for 10 min, the pH was adjusted to 4 with acid solution, and the precipitate was collected by centrifugation at 3300xg for 20 min at room temperature. The precipitate was washed repeatedly with 10 times the weight of distilled water and centrifuged at 4500xg until the washing solution became neutral, and then freeze-dried in vacuo to obtain 182 g of squid ink melanin. After analysis, the purity was 86.2%.

Abstract

The process of preparing melanin of squid ink includes the following steps: taking squid ink, washing with distilled water, filtering with filtering cloth to eliminate impurity, regulating pH value with alkali solution to 5.0-10.0, adding enzyme and hydrolyzing at 35-70 deg.c via stirring for 12-24 hr, heating to 90-100 deg.c to terminal reaction, regulating pH value to 1.0-5.0 with acid, centrifuging at 3000-5000xg and in room temperature to collect precipitate, washing the precipitate with distilled water, centrifuging and vacuum drying. The process has product purity of 30-99 % and extraction rate of 6-19.9 %. The present invention has the advantages of rich material source, high melanin content, simple preparation process, no environmental pollution, etc. The melanin product may be used in preparing health product or medicine for resisting oxidation, regulating immunity, promoting hematopoiesis or lowering blood fat.

Description

Technical field [0001] The invention relates to a preparation method and application of squid ink melanin. Background technique [0002] Black food is popular all over the world. Melanin is the only natural endogenous biopolymer known to protect organisms from radiation damage. It has the functions of anti-oxidation, promoting immune function, promoting bone marrow hematopoiesis and preventing aging. At present, natural melanin mainly comes from the black leaves and fruits of plants, silky fowl and animal hair, and melanin-producing microorganisms. Different sources of melanin have different physiological functions. The preparation process of melanin derived from animals and plants mainly adopts concentrated acid hydrolysis. This method has the disadvantages of severe hydrolysis conditions, easy damage to the morphology and chemical structure of melanin, and low product yield. In particular, with the continuous emergence of diseases such as mad cow disease and avian flu in recent...
