Chicken infectious bursal disease very virulent cell adapted strain and application thereof

A technology for bursal disease and chicken infectivity. It is applied in the direction of antiviral agents, viruses/phages, and viral antigen components. It can solve the problems of virulence and worry about the consequences of use, and achieve the improvement of antigen quality, small immune dose, and protection. high rate effect

[0027] 1.1 Identification of Seed Viruses for Vaccines

[0028] 1.1.1 Cultivation and toxicity determination of a kind of poison

[0029] Chicken embryo fibroblasts were prepared from 10-day-old SPF chicken embryos, inoculated with 5-generation cytotoxicity in the manner of synchronous inoculation, that is, the super-virulent cell-adapted strain IBDV-G of chicken infectious bursal disease of the present invention, and cultured at 37°C . The virus cultures were collected at different times, the virus content was determined by the method of virus plaque formation, the virus growth curve was drawn, and the time of virus collection and the price of seed virus were determined.

Abstract

The invention discloses a chicken infectious bursal disease very virulent cell adapted strain and application thereof in preparation of a drug for preventing chicken infectious bursal disease. The cell adapted strain is the cell adapted strain of a chicken infectious bursal disease very virulent Gx strain, is namely as IBDV-G strain with culture collection number of CGMCC (China General Microbiological Culture Collection Center) 5553. The IBDV-G strain is inactivated and prepared into a vaccine to be used for animal immunization. The results prove that the vaccine is safe, and has 100% protective rate on very vilurent IBDV-G lethal hit; and the antibody positive rate, antibody titer and very virulent protective rate of the very virulent inactivated vaccine are ranked at the top as compared with those of the compared vaccines. Therefore, the popularization and application of the chicken infectious bursal disease very virulent cell adapted strain can effectively prevent the outbreak andprevalence of vvIBD in local area in China and favorably promote sustained, stable and sound development of immune areas or even poultry husbandry in China.

Description

technical field [0001] The invention relates to a cell-adapted strain of a super-virulent strain and its application, in particular to a cell-adapted strain of a super-virulent chicken infectious bursal disease and its use in preparing medicines for preventing chicken infectious bursal disease application. It belongs to the field of biological vaccines. Background technique [0002] Infectious bursal disease (IBD) is an acute, highly contagious disease of young chickens. Its pathogen is chicken infectious bursal disease virus (IBDV). IBDV belongs to the DiRNA virus family, and the avian diRNA virus is a virus. It has two serotypes, serotype I and serotype II. Type I virus is pathogenic to chickens, and is divided into different subtypes, such as classic strains, mutant strains, super-virulent strains and attenuated strains. IBDV tends to invade active isolated cells. In chickens, it mainly invades the precursor B cells in the central immune organ-the bursa of Fabricius,...
