Embodiment 1

[0024] Add 20L of pure water to the reaction kettle, start stirring at 200rpm, heat to 45°C, slowly add 2Kg of casein into the reaction kettle, adjust the pH to 7.0; put in 10g of papain. After 1 hour of enzymatic hydrolysis, the feed solution was cooled to below 30°C, ultrafiltered with a 6000 Dalton roll-type ultrafiltration membrane, concentrated to 4L, and the permeate was collected to a centrifuge collection tank. Add pure water to the concentrated solution to 16L and return to the enzymolysis tank, raise the temperature to 45°C, adjust the pH to 7.0, and continue the enzymolysis. After 1 hour of enzymatic hydrolysis, the temperature of the feed liquid was lowered to below 30°C, concentrated by ultrafiltration to 4L, and the permeate was collected into a centrifuge collection tank. Add pure water to the concentrate again to 16L and return to the enzymatic hydrolysis tank, raise the temperature to 45°C, adjust the pH to 7.0, and continue the enzymatic hydrolysis, and the e...

Abstract

The invention discloses a method for preparing casein phosphopeptide. The method comprises the following steps of: (1) adding protease into casein to perform enzymatic hydrolysis; (2) separating out enzymatic hydrolysate namely casein phosphopeptide by using an ultrafiltration membrane; (3) desalinating the casein phosphopeptide which is produced after ultrafiltration by using a nanofiltration membrane; and (4) adding protease into trapped fluid of the ultrafiltration membrane to perform the enzymatic hydrolysis, and repeating the steps (2) and (3). The method for preparing the casein phosphopeptide is simple to operate, low in equipment requirement, high in yield and high in product quality.

Description

technical field [0001] The invention belongs to the field of biotechnology and relates to a method for preparing casein phosphopeptide. Background technique [0002] Casein phosphopeptides (CPPs) are natural biologically active peptides containing clustered phosphoserine, which are obtained from bovine milk casein through hydrolysis, separation and purification catalyzed by a single or multiple enzymes. It can be used as a carrier of inorganic ions. It can combine with calcium ions, ferrous ions and zinc ions at the lower end of the small intestine of animals with neutral to slightly alkaline pH to prevent the precipitation of metal ions and phosphate, and make the soluble calcium in the small intestine , iron and zinc concentrations are increased, thereby promoting the absorption and utilization of these essential metal ions. In addition, in animal and human experiments, CPPs exhibited anti-caries, anti-bone loss, enhanced fertilization and embryonic development, etc. [...
