Abstract

The invention discloses an inducing method of polyploidy jatropha curcas. The method comprises the following steps of: putting degreasing cotton on a growth point at the top end in the middle of two cotyledons when two cotyledons and a main leaf grow on a jatropha curcas seeding; dripping 0.1-0.8% colchicine solution which is prepared by 1% dimethylsulfoxide (DMSO) by three times everyday on the degreasing cotton; transplanting into a field to grow after the seeding which is lastingly processed for 1-10 days continues to grow for two weeks in a culture room; screening the polyploidy when the height of the plant is 50 cm according to the sizes of pores in lower epidermis of the leaf; further determining the deoxyribonucleic acid (DNA) content in a cell nucleus of a mature leaf to identify the polyploidy by adopting a flow cytometer; and detecting and counting the chromosome number in apical cell nucleus of a germinating seed for the polyploidy jatropha curcas capable of blossoming and bearing fruit normally. The method is simple and convenient, demands on laboratory conditions is low, experimental repeatability is good, inducing cycle is short, and the efficiency is high; the first generation of seed of the polyploidy can be obtained within a year; the method has a good application prospect.
