Problems solved by technology

At present, there is "GB / T18653-2002" standard test method for Campylobacter fetalis in frozen semen of sheep and cattle in China. This method is only suitable for liquid samples and has certain limitations for solid samples; Bacteria culture conditions are harsh and growth is slow, and it takes 7 to 9 days from culture to biochemical identification, which will delay the corresponding diagnosis and treatment; moreover, the operation of campylobacter culture is cumbersome and requires a microaerobic environment, and 8 types of culture are required from cultivation to biochemical identification basis, 10 trials

Abstract

The invention discloses a method and a kit for quickly identifying campylobacter fetus and venereal disease sub type. According to the method, with a genomic DNA of a to-be-detected sample as a template, primers shown in SEQ ID NO. 1, 2, 4 and 5 and probes shown in SEQ ID NO.3 and 6 are added in a qPCR system to perform double real-time fluorescence quantification PCR; if the Ct value is less than 35, the result is positive; if the Ct value is between 35 and 40, the result is suspicious, and another test needs to be carried out; and if the Ct value is greater than 40, the result is negative. The kit using the method mainly comprises the primers and the probes. The method and kit disclosed by the invention have the advantages that the detection time can be greatly shortened, and the integral detection can be finished within 1 day; the method and kit are simple to operate by only extracting nucleic acid and carrying out fluorescent PCR without extra operations; the campylobacter fetus and venereal disease sub type can be quantitatively and qualitatively detected at one time; the kit is high in sensitivity and can provide accurate and reliable results.

Description

technical field [0001] The invention belongs to the field of biotechnology, in particular to a method and a kit for rapidly identifying fetal campylobacter and venereal subspecies. Background technique [0002] Campylobacter fetalis is a kind of Gram-negative, oxidase-positive, catalyst-positive, comma or S-shaped curved, active, microaerophilic zoonotic infectious disease pathogen. Campylobacter fetalis often settles in the intestinal tract or genitourinary tract of wild animals, poultry, livestock and pets, and infects humans through the "fecal-oral" route, often causing human extraintestinal infections, such as bacteremia, meningitis, joint Inflammation, cellulitis, aneurysm infection and other diseases. Campylobacter fetus can be divided into fetal subspecies and venereal subspecies. Venereal subspecies often exist in the reproductive tract of animals and are transmitted through mating and artificial insemination, often causing infertility, abortion and reproductive tra...
