[0025] The preparation method of a kind of probiotic microcapsule that the present invention relates to is realized by the following steps:

[0026] Step 1: Under the temperature condition of 37°C, anaerobically culture the probiotic powder in the liquid medium for 24 hours, collect the bacteria by centrifugation, and wash with 0.85% normal saline to prepare the bacteria suspension;

[0027] Step 2: Prepare a protective agent at a temperature of 78°C. Add 30g of inulin and 0.1g of seabuckthorn powder to each 100mL of the protective agent, and the rest is distilled water;

[0028] Step 3: Cool the sterilized protective agent at room temperature, mix the protective agent and the bacterial suspension evenly at a volume ratio of 3:1, let it stand for 30 minutes, and pre-freeze it at -20°C for 2 hours, then drying in a vacuum freeze-drying oven at -50°C and a vacuum of 50-100 mTorr for 48 hours to prepare freeze-dried bacteria powder;

[0029] Step 4: Add 0.1 g of freeze-dried bac...

Abstract

The invention relates to a probiotics microcapsule and a preparation method thereof. The probiotics is obligate-anaerobic, and cannot keep the activity during production, storage and transportation. The preparation method comprises the following steps: after performing anaerobic culture in liquid culture medium, centrifuging probiotics powder and collecting thalli, and preparing a bacterium suspension; mixing and pre-freezing a protecting agent and the bacterium suspension, and performing vacuum freeze-drying to obtain lyophilized bacterial powder, adding the lyophilized bacterial powder into a sodium alginate solution to obtain a bacteria-gelatin mixed solution; dropwise adding the bacteria-gelatin mixed solution into soybean oil, emulsifying and rapidly adding into calcium chloride solution; and centrifuging and collecting microcapsules. The probiotics microcapsule has the advantages of resisting gastric acid, cholate and enteric solubility, the survival rate of thalli is high, the preparation method is simple, the practicability is strong, and the industrial production is facilitated; and as fructooligosaccharide, inulin in the protecting agent not only can be proved to improve the survival rate of probiotics.

Description

technical field [0001] The invention belongs to the technical field of probiotic product preparation, and in particular relates to a probiotic microcapsule and a preparation method thereof. Background technique [0002] Probiotics are a class of active microorganisms that are beneficial to the host. They are a general term for active beneficial microorganisms that colonize the human intestinal tract and reproductive system and can produce definite health effects, thereby improving the host's micro-ecological balance and exerting beneficial effects. Common probiotics include Bifidobacterium longum, Lactobacillus acidophilus, Lactobacillus rhamnosus and Bifidobacterium lactis. They are all important probiotics in human and animal intestines, which have the properties of regulating the balance of intestinal flora, providing nutrition, lowering cholesterol and anti-cancer. However, they are obligate anaerobic and are extremely sensitive to external adverse environments such as ...
