Problems solved by technology

However, this method has the following problems: 1) The test cycle is too long and the throughput is small: According to the plant pictures of this method, the plants should grow for about 35-40 days, and the long test cycle leads to low identification efficiency and low throughput, which cannot meet More samples need to be identified; 2) The required cost is higher: the flower pots used in this invention are larger, resulting in the doubling of the costs of the cultivation substrate, nutrient solution, and space required for the test in the later stage. It may not be possible to carry out experiments at all under conditions of stable factors such as a climate incubator
However, in this method, the assembly of equipment is relatively cumbersome, and there is a problem of inconsistent salt concentration in the sand culture medium.
[0007] In addition to the deficiencies mentioned above, the identification methods disclosed above also have the problem of excessive data processing

Embodiment 1

[0036] With 39 maize inbred lines as the research object, the salt tolerance in seedling stage was identified. The types of materials include waxy corn and ordinary corn, and they are sourced from tropical regions, the United States, and Huanghuaihai. The specific implementation steps are:

[0037] 1) Seed selection

[0038] Choose complete, full, and uniform corn seeds from each inbred line;

[0039] 2) Seed disinfection and nursery

[0040] After disinfection with 10g / L sodium hypochlorite, the seeds are evenly sown in a plastic dish paved with vermiculite, moistened with water, covered with plastic wrap, and cultured in the dark in an incubator at 28°C; 3 days later, wait until the corn seeds have grown roots. Remove the fresh-keeping film and move to an artificial climate room for cultivation. The cultivation conditions are: 35% constant humidity, light time 14h / day, light 5000LX, temperature 25℃ with light, and 10℃ without light. The test material list is shown in Table 1:

Embodiment 2

[0057] According to the salt tolerance parameters of the 39 inbred lines mentioned above, an evaluation system of corn seedling salt tolerance was established.

[0058] 1) Calculation of the salt tolerance coefficient of each trait

[0059] Substitute the above data into the following formula to obtain the salt tolerance coefficient of each index except the salt tolerance level:

[0060] Salt tolerance coefficient (%)=measured value of test group / measured value of control group×100%

[0061] Among them, the measured value of the test group is the test group C, PH, RH, YF, GF, YD, GD obtained by the measurement, and the measured value of the control group is the control group C, PH, RH, YF, GF, YD, GD obtained by the measurement;

[0062] The results are shown in Table 3;

[0063] Table 3 Salt tolerance coefficient of various properties of the tested materials

Abstract

The invention belongs to the technical field of crop planting, and particularly relates to an authentication and evaluation method for the salt tolerance of corns in a seedling stage. The authentication method comprises the following steps: (1) selecting, disinfecting and culturing seeds, and culturing seedlings to a three-leaf one-core stage; (2) dividing the three-leaf one-core stage seedlings with the consistent growth vigor into two parts: one as a test group and one as a contrast group, wherein a nutritional solution for the test group contains 150 to 200 mmol / L NaCl; (3) sucking water ofa plant with water sucking paper till the plant is dry, measuring chlorophyll contents (C) of third leaves of the test group and the contrast group, measuring the greatest length from the root part to the leaf tip, recording the greatest length as a plant height (PH), measuring the length of the longest seedling, recording the length as a root length (RH), recording a fresh weight above the rootpart as a leaf fresh weight (YF), recording a fresh weight of the root part as a root fresh weight (GF), then treating the leaf part and the root part at 120 DEG C for 20 min and at 80 DEG C for 48 h,weighing dry weights, and respectively recording the dry weights as a leaf dry weight (YD) and a root dry weight (GD); and (4) substituting the values into the following formula, and calculating a salt tolerance coefficient of each character: salt tolerance coefficient= measured value of the test group / measured value of the contrast group. The method increases the flux, realizes accurate test, shortens the test period, improves the working efficiency, effectively processes data and accurately evaluates the salt tolerance.

Description

Technical field [0001] The invention belongs to the technical field of crop planting, and specifically relates to an identification and evaluation method for salt tolerance at the seedling stage of corn. Background technique [0002] Corn is my country's largest food crop, with the characteristics of great potential for increasing production and diversified uses. With the changes in people's consumption structure, the demand for meat, eggs, milk and other livestock and poultry products is increasing, and the market demand for corn continues to grow. In terms of planting area, the existing fertile land has been fully utilized. There is little potential for increasing planting area by competing with other crops or increasing the multiple cropping index. Fully developing and using saline land to expand planting area is an effective way to increase corn yield. There are about 100 million hectares of saline-alkali land in my country. Only the saline-alkali land area in the Bohai Rim ...
