Embodiment

[0024] The process for extracting heparinoids from heparin by-products in this embodiment comprises the following steps:

[0025] (1) Dissolve 80kg of heparin by-products in 1000kg of hot water, then add 50kg of analytically pure sodium chloride with a mass percentage of 5% until completely dissolved;

[0026] (2) After 80 kg of heparin by-products are completely dissolved, add 3 kg of enzyme reagent, which is made of 14 parts of cysteine, 2 parts of trypsin and 0.5 part of sodium sulfite in parts by weight, and adjust the pH to 9; then heat to 55°C and keep it warm for 2 hours, then raise the temperature to 100°C, and finally cool down to room temperature with circulating water. After cooling down to room temperature, the first centrifugation removes solid impurities to obtain 1200kg of first centrifugate;

[0027] (3) Add 12kg of precipitant to the first centrifugal liquid, which is made of 1.5 parts by weight of 5-sulfosalicylic acid, 22.5 parts of acetone, 57.5 parts of me...

Abstract

The invention relates to a process for extracting heparinoid or protein from heparin by-products. The method comprises the following steps: (1) adding hot water into the heparin by-products for dissolving, and then adding sodium chloride until the heparin by-products are completely dissolved; (2) after the heparin by-products are completely dissolved, adding an enzyme reagent A, adjusting the pH to 8-10, heating and then cooling to the room temperature, and centrifuging to obtain first centrifugate; (3) adding a precipitant B into the first centrifugate, stirring, and centrifuging to obtain second centrifuged wet solid; (4) adding hot water into the second centrifuged wet solid for dissolving, and then adding the sodium chloride until the solid is completely dissolved; (5) after the secondcentrifuged wet solid is completely dissolved, adding ethanol for precipitating, and collecting precipitate I; (6) adding water for dissolving the precipitate I, and carrying out ultrafiltration to obtain ultrafiltrate; (7) adding the sodium chloride into the ultrafiltrate for dissolving, then adding ethanol, and collecting precipitate II; (8) drying the precipitate II to obtain the heparinoid orthe protein. The process has the advantages that the process can improve the recovery rate and titer of the heparinoid or the protein.

Description

technical field [0001] The invention relates to a preparation process of heparin, in particular to a process for extracting heparin or protein from heparin by-products. Background technique [0002] Heparin, another name protein, refers to natural, synthetic and semi-synthetic substances that have similar effects to heparin. Heparin can combine with hundreds of proteins and affect the spatial conformation, lifespan and biological activity of proteins. Heparin also It can be combined with a variety of proteins in the extracellular matrix to control their distribution, migration and information transmission. It can be seen that heparinoid is an important information molecule, which regulates the behavior of host cells, such as cell proliferation, differentiation, and combination, and participates in the process of identifying and controlling foreign cells. Heparin-like products can be used as raw materials for the production of sulodexide, Guanxinshu and other drugs. These dr...
