[0066] 1.1 Instruments and reagents

[0067] The oligonucleotides in this example were synthesized and purified by Shanghai Sangon Biotechnology Co., Ltd., and their sequences are listed in Table 1. Tetraethoxysilane (TEOS) was obtained from Shanghai Sinopharm Co., Ltd. Cetyltrimethylammonium nitrate (CTAB) and epigallocatechin-3-gallate were purchased from Solebo Bioscience and Technology Ltd. Coumarin 6 and glutathione were purchased from Aladdin Biochemical Technology Co., Ltd. Adriamycin was purchased from Huafeng Group Co., Ltd. Paclitaxel was provided by Chenxin Pharmaceutical Co., Ltd. MTT was purchased from Sigma-Aldrich Corporation. All solutions were prepared with ultrapure water (>18.25MΩ). All other chemicals used in the experiments were of analytical grade and used without further purification.

[0068] Fluorescence measurements were performed on a Hitachi F-7000 fluorescence spectrophotometer. Ultraviolet absorption spectrum scanning was performed using a ...

Abstract

The invention belongs to the technical field of nano-drug delivery systems, and specifically relates to a subcellular targeted nano-drug delivery system. For problems in the prior art that an antitumor drug synergistic delivery system is hard to realize subcellular targeting, an MSN synergistic delivery nanocarrier whose assemblies in cells are detachable can be provided; the nanocarrier is used for the drug delivery of subcellular targeting; the carrier is in a core-shell-hook structure, and adopts MSN as a nuclear material to carry PTX; polymerized polyphenol (EGCg) can be taken as a shell material to package the drugs in MSN (MSN@EGCg) and provide modification sites for a hook; and aptamer AS1411 linked double-stranded DNA (dsDNA-Apt) can be taken as the hook to combine nucleolin and carry Dox. The nanocarrier can respectively deliver the Dox and PTX to nucleuses and cytoplasm, so that intracellular targeting ability of drug delivery can be enhanced; and thus, the system can be applied to the development of antitumor drugs, and has considerable prospects.

Description

technical field [0001] The disclosure belongs to the technical field of nano drug delivery system, in particular relates to a nano drug delivery system with core-shell-hook structure and subcellular targeting. Background technique [0002] The information disclosed in this Background section is only intended to increase the understanding of the general background of the disclosure, and is not necessarily to be taken as an acknowledgment or any form of suggestion that the information constitutes prior art that is already known to those skilled in the art. [0003] Although chemotherapy is a classic means of cancer treatment, its efficacy is often limited by the non-selective delivery of free chemotherapeutic drugs. These drugs indiscriminately enter normal cells and cancer cells by means of simple diffusion, inevitably resulting in decreased chemotherapeutic efficacy and severe side effects. In order to solve these problems, researchers in this field have found that nanopart...
