Problems solved by technology

Chemical fungicides have been widely used for a long time because of their advantages of high efficiency, low price, and convenient use. However, this method has disadvantages such as causing drug resistance of pathogenic bacteria, endangering human health, and polluting the environment.
However, there is no safe and effective biological control method for reducing postharvest diseases of asparagus at present, and there is no research report on the use of Yarrowia lipolytica to control postharvest diseases of asparagus

Embodiment 1

[0032] Screening for the concentration of Yarrowia lipolytica WSL-01 used to control the postharvest disease of asparagus;

[0033] 1. Test plan;

[0034]The tested asparagus had uniform thickness, consistent color, and no mechanical damage or pest damage on the surface. First rinse the soil on the surface of the asparagus with clean water, then soak it in 3% sodium hypochlorite solution for 5 minutes, rinse it with sterile water, and dry it at room temperature. Make a wound with a hole diameter of 3 mm and a depth of 2 mm on the tip and base of the asparagus respectively; add 20 μL of the following treatment solution to different wounds: (1) 1×10 6 cells / mL of Yarrowia lipolytica WSL-01 suspension; (2) 1×10 7 cells / mL of Yarrowia lipolytica WSL-01 suspension; (3) 1×10 8 cells / mL of Yarrowia lipolytica WSL-01 suspension; (4) 1×10 9 cells / mL of Yarrowia lipolytica WSL-01 suspension; (5) sterile saline;

[0035] After standing for 2 h, inoculate 20 μL of 5×10 4 spores / mL p...

Embodiment 2

[0042] Controlling effect of Yarrowia lipolytica WSL-01 on postharvest natural rot of asparagus;

[0043] 1. Test plan;

[0044] The tested asparagus had uniform thickness, consistent color, and no mechanical damage or pest damage on the surface. Spray 1×10 evenly on the surface of unwashed and sanitized asparagus 7 cells / mL of Yarrowia lipolytica WSL-01 suspension, and asparagus sprayed with sterile saline as a control group. After natural drying, the asparagus was sealed with plastic wrap in a plastic basket, and placed in a constant temperature and humidity incubator (humidity 90%) at 20°C for storage. Each treatment was replicated three times with 16 asparagus per replicate. The whole experiment was repeated 2 times.

[0045] The formula for calculating the disease index is as follows:

[0046] Disease index = ∑ (number of diseased plants at all levels × representative value of relative series) / (total number of plants × representative value of the highest level)

Embodiment 3

[0050] Effects of Yarrowia lipolytica WSL-01 on the content of soluble sugar, vitamin C and chlorophyll in postharvest asparagus;

[0051] 1. Test plan;

[0052] The tested asparagus had uniform thickness, consistent color, and no mechanical damage or pest damage on the surface. Spray the following treatment solution evenly on the surface of unwashed and disinfected asparagus: (1) 1×10 7 cells / mL of Yarrowia lipolytica suspension; (2) the control group was sterile saline. After natural drying, the processed asparagus was sealed in a plastic basket with a plastic wrap, and placed in a constant temperature and humidity incubator (humidity 90%) at 20°C for storage. Each treatment was replicated three times with 16 asparagus per replicate. The whole experiment was repeated 2 times.

Abstract

The invention belongs to the field of biological control of postharvest diseases, and specifically relates to application of Yarrowia lipolytica in postharvest disease control of asparagus and use ofstorage and preservation. The steps are as follows: activating and cultivating Yarrowia lipolytica, conducting centrifugation to obtain bacterial cells, washing the bacterial cells with sterile physiological saline, conducting centrifugation, diluting the obtained bacterial cells with sterile physiological saline to a bacterial suspension at a concentration of 1x10<7> cells / mL, spraying the bacterial suspension directly and evenly on the surface of asparagus, and conducting air-drying naturally; and post-harvest disease control, storage and preservation of asparagus can be realized. Accordingto the present invention, root rot disease and natural rot of postharvest asparagus can be effectively controlled by Yarrowia lipolytica, so as to reduce the loss caused by the postharvest diseases ofasparagus; and at the same time, Yarrowia lipolytica has no adverse effects on the content of soluble sugar, vitamin C and chlorophyll which are the main quality indexes of postharvest asparagus, issafe and environment-friendly, can replace chemical bactericides to control the postharvest diseases of asparagus, and can reduce the potential harm of the chemical bactericides to human bodies and environment.

Description

technical field [0001] The invention belongs to the field of biological prevention and control of postharvest diseases of vegetables, and in particular relates to the use of Yarrowialipolytica in the prevention and control of asparagus postharvest diseases and storage and preservation. Background technique [0002] Asparagus, scientific name Asparagus officinalis, also known as Asparagus officinalis, is a perennial herb of the genus Asparagus in the family Liliaceae. Green asparagus contains chlorophyll, amino acids, multivitamins, carbohydrates, calcium, phosphorus and other minerals and various active ingredients, and has high nutritional value. It has curative effect on many diseases such as hypertension, hyperlipidemia, arteriosclerosis, heart disease, hepatitis, cirrhosis, nephritis, edema, and cystitis. Long-term consumption can help digestion, enhance appetite, improve body immunity, reduce the toxicity of harmful substances, inhibit the vitality of cancer cells, pre...
