Abstract

An exosome preparation includes exosome secreted by human umbilical cord mesenchymalstem cells and PBS. A preparation method of the exosome secreted by the human umbilical cord mesenchymal stem cells includes placing the umbilical cord mesenchymal stem cells separated from the umbilical cord of a full-term normal delivered healthy fetus to perform culture at 37 DEG C to P3; performing replacing in a DMEM / HAM'SF-12 culture medium without fetal bovine serum when the cells are fused to 60-80%, and performing starvation culture for 48 h, and then collecting the culture medium; performing centrifugation on the collected culture medium to remove the cells and cell debris, and extracting the exosome by using a ultracentrifugation method; and obtaining the mesenchymal stem cell exosome preparation UC-MSCs-Exo by performing conservation at -80 DEG C with a PBS re-suspended sediment. The UC-MSCs is low in preparation cost, abundant in source and convenient in collection and has no traumatic operation on human bodies; the UC-MSCs-Exo is simple in component, stable in cell factor and not easy to denature; and therefore, the skin tissue structures at alopecia areata parts can be improved, and the damaged hair follicles can be repaired or reconstructed.
