Abstract

The method comprises the following steps: 1) selecting 9-11 plant roots which need to be subjected to vitality determination and have the length of 1 cm or integral multiples of 1 cm, cutting the plant roots into sections in the length of 1cm, weighing root sections on a balance, and putting the root sections with the weight range of 20-30 mg into a centrifuge tube filled with 1 ml of TTC; 2) putting the centrifuge tube into a water bath kettle at 40 DEG C, incubating the tube for 30 minutes, and adding 1ml of sulfuric acid into the centrifuge tube to stop reaction; 3) sucking out a mixed liquid of TTC and sulfuric acid in the centrifuge tube, rinsing the root section with deionized water to removeresidual liquid on the surface of the root section, cutting the root section into pieces with scissors, and adding 1 ml of 95% ethanol for immersion extraction; and 4) absorbing the ethanol extracting solution in the centrifugal tube, measuring the light absorption value at 485nm by using a spectrophotometer, and representing the root activity of different root sections of the plant root system by using the light absorption value per unit weight.
