Problems solved by technology

However, the inventors found that during the long periodontitis clinical treatment period, due to the lack of plasticity of the bracket material, the poor adaptability and the inability to fully fill the periodontal defect, the patient's comfort and compliance with treatment often decrease, resulting in Poor or even failure of treatment

[0041]In another specific embodiment of the present invention, there is provided a preparation method of the above-mentioned targeted antibacterial and in-situ bone formation dual-functional material. The preparation method comprises: combining polyethylene glycol diacrylate, SDF-1 and functional polypeptide module Mix well and get it.

[0042]In another specific embodiment of the present invention, the preparation method adopts a secondary mixing method, that is, after mixing polyethylene glycol diacrylate and SDF-1, the functional polypeptide module is added for mixing. The ethylene glycol diacrylate and the functional peptide module undergo a Michael addition reaction and form a temperature-sensitive gel. Tests have proved that the gel solution is liquid at room temperature (25°C). When transferred to an environment of 37°C, it can automatically crosslink into a hydrogel state within 30 minutes.

[0060]1. Development and preparation of functional peptide module: Using solid-phase peptide synthesis, a peptide chain with a total length of 30 amino acids was synthesized de novo. The amino acid sequence of the peptide chain is: CGPQRIWGQCGGVVFGVGFGCGPQRIWGQC (SEQ ID NO. 1). Preparation of polyethylene glycol diacrylate: In a three-necked bottle equipped with a water separator, heat 0.1 mol of PEG20000 (the polymerization inhibitor is 0.3 g of hydroquinone) to 70 degrees Celsius, and then add 0.3 mol of acrylic acid and 4 g of p-toluenesulfonic acid were stirred. After the temperature was raised to 110 degrees Celsius, the reflux reaction was maintained for 2 hours, and then the temperature was raised to 120 degrees Celsius until ...

Abstract

The invention provides a targeted antibiosis and in-situ osteogenesis promoting bifunctional material as well as a preparation method and application thereof, and belongs to the technical field of periodontal tissue repair and regeneration. The bifunctional material comprises a gel scaffold and SDF-1, wherein the gel scaffold is formed by cross-linking PEG-DA and Peptide, has a temperature-sensitive characteristic and can respond to gingipains, and the SDF-1 is further loaded by the material and has a recruited stem cell homing function. By developing and preparing the gingipain response typetemperature-sensitive gel material, long-acting targeted antibiosis and in-situ regeneration promotion of a periodontal defect part can be realized simultaneously along with release of short antibacterial peptide and the SDF-1. Tests prove that the PEG-DA-Peptide@SDF-1 bifunctional material prepared by the invention is temperature-sensitive and can be applied by injection, and has good long-actingtargeted antibiosis and in-situ regeneration promoting effects, so that the bifunctional material has good practical application value.

Description

Technical field[0001]The invention belongs to the technical field of periodontal tissue repair and regeneration, and specifically relates to a targeted antibacterial and in-situ bone-promoting dual-function material, and a preparation method and application thereof. Background technique[0002]Disclosure of the background information is only intended to increase the understanding of the overall background of the present invention, and is not necessarily regarded as an acknowledgement or in any form suggesting that the information constitutes the prior art known to those of ordinary skill in the art.[0003]Periodontitis is a periodontal disease caused by the infection of various anaerobic pathogens such as Porphyromonasgingivalis (P.gingivalis). The alveolar bone resorption caused by it is considered to be adult loss of teeth The main reason. Periodontitis not only causes bleeding, ulceration and loose teeth in the gums, but also has a close relationship with systemic diseases such as di...
