Embodiment 1

[0032] Example 1 Isolation, identification and preservation of strains

[0033] 1. Isolation and purification of strains

[0034] Using different kinds of kimchi as the separation source, take the samples for 10-fold serial dilution with normal saline, draw 100 μL of the diluted solution and spread it on MRS agar medium, cultivate at 37 °C for 48 hours, take a single acid-producing colony for purification by partition and continuous purification More than three generations of a single colony (see figure 1 ).

[0035] 2. Identification of strain BYL4.2

[0036] The purified single colonies were picked for Gram staining and microscopic examination, and the morphology of the cells was observed under a light microscope. Strain BYL4.2 is rod-shaped, Gram-positive (see figure 2 ), which were then molecularly identified based on 16S rDNA sequencing. DNA was extracted from the bacterial culture using the Ezup Column Bacterial Genomic DNA Extraction Kit. The primers for amplifyi...

Embodiment 2

[0041] Embodiment 2 The performance verification of Weissella sinensis BYL4.2

[0042] 1. The growth curve and pH change curve of Weisseria sinonaea BYL4.2

[0043] The strain BYL4.2 was inoculated into MRS broth with 1% inoculum, and cultured at 37°C for 24h, and the OD was determined by sampling every 2h. 600nm and pH value, to detect the growth change and pH change of strain BYL4.2 within 24h, the results are as follows Figure 4 . It can be seen that the lag period of strain BYL4.2 is short, entering the logarithmic growth phase in 2h, and entering the stable phase in about 12h. The pH value changed greatly in the logarithmic growth phase, and the pH value dropped to about 4.5 after entering the stable phase for 12 hours.

[0044] 2. Antifungal activity of Weisseria sinonaea BYL4.2

[0045] Penicillium chrysogenum and Aspergillus versicolor were inoculated on PDA solid medium and cultured at 28°C for 3 days until a large number of spores grew on the medium. Add a smal...

Abstract

The invention discloses Weissella sinus with broad-spectrum antibacterial activity, and belongs to the field of microorganisms. The strain is preserved in the China Center for Type Culture Collection, the preservation number is CCTCC NO: M 20211696, the preservation address is Wuhan University, Wuhan, China, and the preservation date is December 29, 2021. The Weissella cibaria BYL4.2 disclosed by the invention is separated from pickles, and experiments prove that the Weissella cibaria BYL4.2 can inhibit growth and reproduction of penicillium chrysogenum, aspergillus versicolor, escherichia coli, staphylococcus aureus, listeria monocytogenes and pseudomonas aeruginosa in vitro, and has broad-spectrum antifungal and bacterial activity; moreover, the compound is sensitive to antibiotics such as erythromycin and rifampicin, is relatively high in safety, is high in survival rate in gastric acid and intestinal high-bile-salt environments, and has potential application prospects in the fields of improvement of the intestinal environment and preservation and fresh keeping of foods.

Description

technical field [0001] The present invention relates to the field of microorganisms, in particular to a strain of Weisseria sinonaea having broad-spectrum antibacterial activity. Background technique [0002] In the prior art, in order to delay the deterioration rate of food and improve the shelf life of the product, the usual method is to add chemical preservatives. However, this method is not in line with the theme of green, environmental protection and sustainable development advocated by the state. Moreover, long-term consumption of food containing chemical preservatives is likely to cause disturbance of the intestinal flora of consumers, thereby affecting their health. [0003] Lactic acid bacteria are recognized as food-grade safe microorganisms. During the fermentation of carbohydrates, metabolites include proteins, polypeptides, organic acids and other substances with bacteriostatic activity. These bacteriostatic substances can inhibit the growth and reproduction of...
