Climbazole is a topical antifungal agent commonly used in the treatment of human fungal skin infections such as dandruff and eczema. Climbazole has shown a high in vitro and in vivo efficacy against Pityrosporum ovale that appears to play an important role in the pathogenesis of dandruff. Its chemical structure and properties are similar to other fungicides such as ketoconazole and miconazole.
Preparation method of high-purity climbazole
The invention discloses a preparation method of high-purity climbazole, and relates to the technical field of synthetic chemistry. The preparation method of the climbazole comprises the following steps: reacting pinacolone with chlorine to obtain chloropinacolone, reacting the chloropinacolone with parachlorophenol to obtain ether ketone, chlorinating the ether ketone with a chlorinating agent to obtain chlorinated ether ketone, reacting the chlorinated ether ketone with imidazole to obtain a crude climbazole product, and refining the crude climbazole product with activated carbon, filtering, crystallizing, centrifuging and drying to obtain the climbazole. The chlorination reagent used in the ether ketone chlorination stage and a proper adding mode improve the reaction selectivity, SO2 by-products which cause pollution to the environment are not generated, and the environmental protection property is high; an additional catalyst is not used in the reaction process of chloro-ether-ketone and imidazole, and recyclable activated carbon is used in the purification of a climbazole crude product, so that the preparation cost is reduced; the climbazole prepared by the method has high yield and high purity, and is suitable for industrial production.
