Embodiment 1

[0027] Gelatin matrices were prepared as follows. The water at the bottom of the double evaporator is heated with a hot plate. Put purified water in a second pan and bring to a boil; part of the water is added to the upper part of the double evaporator. Sprinkle in the gelatin and stir gently to hydrate for about 30 minutes. Add purified water, sodium benzoate and starch into a sauce pan, stir the starch to form a slurry. Corn syrup (dextrose equivalent 43) was added to the starch slurry. Heat the starch / corn syrup to a boil with constant stirring. Boiling state by adding sucrose. When the starch / corn syrup is cooked to the desired percent solids, the mixture is cooled to a suitable temperature, the gelatin solution is added and mixed well. Next, the following ingredients were added to the sugar / starch / gelatin mixture: anhydrous citric acid, FD&C Red No. 40., synthetic saccharin (Hoescht-Celanese), and encapsulated acetaminophen. Stir these materials thoroughly to incorp...

Embodiment 2

[0098] The following compositions substitute other hydrocolloids for starch. Samples were prepared in a manner similar to that described in Example 1 above.

[0099] A) Agar 2.87% Gelatin / hydrocolloid ratio 1.00:0.73

[0100] Gelatin 3.93%

[0101] Solid sugar 63.36%

[0102] Encapsulated Acetaminophen 9.25%

[0103] Add remaining solids to 80% w / w during deposition

[0104] Evaluation: 11 seconds on average, hard chewing, very short chewing, very thorough, helpful for mold peeling, brittle, dry, hard, the pieces are completely broken after chewing, and the taste is unpleasant (at this concentration)

[0105] B) HPMC 5.72% gelatin / hydrocolloid ratio 1.00:1.46

[0106] Gelatin 3.92%

[0107] Solid sugar 60.53%

[0108] Encapsulated Acetaminophen 9.25%

[0109] Add remaining solids to 80% w / w during deposition

[0110] Evaluation: The composition was very sticky and could not be peeled from the mold.

[0111] C) microcrystalline cellulose 2.91% gelatin / hydr...

Abstract

The invention relates to a gelatin matrix containing pharmaceutically active ingredients. In a preferred embodiment, hydrocolloids are added to the gelatin matrix to improve the physical properties, taste and mouthfeel of the gelatin matrix. A method of making a gelatin matrix comprising a hydrocolloid is also disclosed.

Description

technical field [0001] The present invention relates to a soft and chewable pharmaceutical dosage form. More specifically, the present invention relates to an oral delivery system for delivering drugs, medicaments or other active ingredients by using a smooth chewable base to mix with the active ingredients. Background technique [0002] Typical dosage forms of pharmaceuticals and nutritional supplements for oral administration are solid forms of tablets, capsules, pills, lozenges or powders. Tablets are swallowed whole, chewed in the mouth, or sublingual. Absorption of the active moiety depends on its release from the dosage form and can be controlled by several different techniques. [0003] Chewable systems are often used for drug delivery when providing a tablet to swallow whole is not practical. The chewing action acts like a tablet disintegrant to help the tablet disintegrate and increase the rate of absorption in the digestive tract. Masticatory systems also have ...
