Problems solved by technology

[0004] However, the bacteria has not been widely used in production. The main reason is that the in vitro culture technology is still immature, and it mainly relies on living hosts for reproduction, so it is difficult to produce on a large scale.

[0018] 1. The regulation of temperature on the development of Pasteurella puncturee

[0019] The regulation of temperature on the growth of Pasteurella puncture is carried out in the following steps:

[0020] 1. Test nematodes and their isolation

[0021] The nematodes tested were root-knot nematode Meloidogyne spp. (China Forestry Microorganism Culture Collection and Management Center). Take the tomato plants infected with root-knot nematode, wash the tomato roots with clean water, place them in 0.5% sodium hypochlorite solution for 30 sec, scrub quickly, sieve, rinse, and collect the eggs of root-knot nematode on a 3-layer filter paper pad. In the nematode separator, hatch at room temperature for 3-4 days to obtain the second instar larvae of root-knot nematode.

Embodiment 3

[0085] Embodiment 3, the regulation and control of temperature to Pasteuria nishizawae development

[0086] 1. The regulation of temperature on the development of Pasteuria nishizawae

[0087] The regulation of temperature on Pasteuria nishizawae is carried out according to the following steps:

[0088] 1. Test nematodes and their isolation

[0089] The nematodes tested were cyst nematodes (Heterodera spp.,) (China Forestry Microorganism Culture Collection and Management Center). Take soybean plants infected with cyst nematodes, pick fresh cysts of cyst nematodes on soybean roots under a stereoscope, select fresh, plump, mature cysts and oocysts, and sterilize them in 0.5% NaOCl solution for 3 minutes, sterile Rinse with water three times, then incubate in sterile water at 25°C, and collect newly hatched cyst nematodes in time every day.

Abstract

The invention discloses a method for controlling the growth and reproduction of pasteurella and use thereof. In the method, pasteurella is cultured at a temperature of equal to or greater than 8 DEG Cand less than 20 DEG C. The invention controls the growth of pasteurella by adjusting the temperature so as to overcome the problem of difficult and complicated obtaining of mycelium through in vitroculture, thereby ensuring the smooth development of in vitro culture in the aspect of material so as to save large amount of workload for preparation of inoculating material and effectively solve theproblem of large space needed by the inoculating material for reproducing living bodies. By using the method in the invention, the pasteurella can be preserved for about 30 days, and after the pasteurella regulated through this method is transferred to the normal condition, the activity of the mycelium does not change; and when the effective accumulated temperature reaches 400 DEG C, the myceliumcan grow to form spores without affecting the yield and quality of the spores. The invention is also applicable to controlling the growth and preserving other pasteurella of other strains, and also applicable to controlling the growth and preserving the pasteurella in artificial culture medium.

Description

technical field [0001] The invention relates to a method for regulating the development and reproduction of bacteria, in particular to a method for regulating the development and reproduction of Pasteurella. Background technique [0002] Pasteuria spp. is a class of parasitic plant-parasitic nematodes, bacteria that can produce hyphae and endospores, and belongs to the low GC content Gram-positive bacteria branch of eubacteria. Pasteurella is an obligate parasite of nematodes, completes its growth and reproduction process in nematodes, and is released back into the soil in the form of spores. Endospores are extremely resistant to stress in the soil. When the host nematodes pass by, the endospores attach to the nematode body wall, germinate as the nematodes develop and invade into the nematode false body cavity, absorbing their nutrients , Destroy its tissue cells, and eventually lead to slow action of nematode larvae and weakened infectivity, or reduce female fertility, the...
